    Transcription factor that is involved in embryonic development, establishment of tissue-specific gene expression and regulation of gene expression in differentiated tissues. Is thought to act as a 'pioneer' factor opening the compacted chromatin for other proteins through interactions with nucleosomal core histones and thereby replacing linker histones at target enhancer and/or promoter sites. Binds DNA with the consensus sequence 5'-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3' (By similarity). In embryonic development is required for notochord formation. Involved in the development of multiple endoderm-derived organ systems such as the liver, pancreas and lungs; FOXA1 and FOXA2 seem to have at least in part redundant roles. Originally discribed as a transcription activator for a number of liver genes such as AFP, albumin, tyrosine aminotransferase, PEPCK, etc. Interacts with the cis-acting regulatory regions of these genes. Involved in glucose homeostasis; regulates the expression of genes important for glucose sensing in pancreatic beta-cells and glucose homeostasis. Involved in regulation of fat metabolism. Binds to fibrinogen beta promoter and is involved in IL6-induced fibrinogen beta transcriptional activation.

    Overlay histogram showing HepG2 cells stained with ab60721 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab60721, 0.5µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

Permeabilierzung: 1% triton x-100 in PBS fuer 1 min, danach Blockierung und Antikoerperfaerbung in 0.1% triton X100 /PBS.
Konzentration der AB69721 10 ug/ml wie bei Ihren Abbildung an dem Datasheet

Keine Insulin ist im Medium. Normales DMEM fuer die Hela Zellen.

Eine Faerbung in der Neuriten von unseren Primaerzellen ist nicht nur eine zytoplasmatische Faerbung. Es ist absolut keine Faerbung im Kern zu sehen.

Ich bitte die Zusendung einen Ersatz-Antikoerper.

wir haben bei Ihnen das Produkt Nr. AB60721
bestellt und auch bekommen (lot #GR 49290-2).
Laut Datenblatt (und mehreren wissenschaftlichen
Artikeln) ist das Epitope im Kern (siehe
Abbildung auf Ihrer Webseite).
Wie Sie an den Bildern im Anhang sehen können,
gibt Ihr Produkt eine starke Cytoplasma- und
Membran-Fluoreszenz an unseren Primärzellen und
auch an Hela Zellen. Es ist sehr stark und
inakzeptabel. Unser zweiter Antikörper hat kein
Signal ohne vorherige Antikörper-Zugabe.
Wir brauchen einen spezifischen anti-Foxa2 Maus
monoklonalen Antikörper für Immunofluoreszenz an
PFA fixierten Zellen.
Bitte ersetzen Sie kostenlos das Produkt AB60721
durch einen entsprechendenen anti-Foxa2 Maus
monoklonalen Antikörper.

Um die Ursache des Problems besser zu verstehen, bitte ich Sie mir die folgenden Fragen noch zu beantworten. Vielen Dank für Ihre Kooperation.

1.) Wurden die Zellen vorbehandelt? FOXA2 kann auch im Zytoplasma sein. Durch Insulinstimulierung zum Beispiel, wird FOXA2 aus dem Nukleus exportiert. http://www.uniprot.org/uniprot/P35583

2.) Wie wurden die Zellen fixiert und eventuell permeabilisiert? Die Fixierung mit Paraformaldehyde funktioniert durch krosslinken, während die Fixierung mit Aceton/Methanol durch Präzipitation funktioniert. In der Tat lohnt es sich deshalb, verschieden Fixierungsmethoden auszuprobieren. 3.) Was für Verdünnungen des ab60721 wurden eingesetzt? War das Resultat anders mit einer hohen Verdünnung?

Vielen Dank für Ihren Anruf.

Wie versprochen habe ich alle unsere FOXA2-Antikörper darauf geprüft, welche Isoformen von FOXA2 sie erkennen. Leider sind alle Immunogene in Regionen lokalisiert, die in beiden Isoformen vorkommen (Details siehe jeweiliges Datenblatt), und daher detektieren die Antikörper immer beide Isoformen. Wir haben also leider keinen Antikörper erhältlich, der zwischen den Isoformen unterscheiden kann.

Es tut mir leid, dass ich keine besseren Nachrichten für Sie habe. Ich hoffe, diese Informationen helfen Ihnen dennoch weiter. Bitte zögern Sie nicht, sich wieder bei uns zu melden, falls Sie weitere Fragen haben.
