Flow cytometry overlay histogram showing HeLa cells stained with ab119688 (red line). The cells were fixed with 4% formaldehyde (10 min) and then permeabilised with 90% methanol for 30 min. The cells were incubated in 1x PBS containing 10% normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab119688) (1x 106 in 100μl at 1.0μg/ml) for 30min at 22°C.

Isotype control antibody (black line) was ab314952 (1.0μg/ml (1/500)) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.

Acquisition of >5000 events were collected using a 50 mW Blue laser (488nm) and 525/40 bandpass filter.

Mouse IgG1 also known as immunoglobulin G1 is an important antibody isotype in mice. It has a molecular mass of approximately 150 kDa. Mouse IgG1 is found in the serum and extracellular fluids where it helps to mediate immune functions. This antibody is produced by plasma cells which originate from B lymphocytes. Its structure is comprised of two heavy chains and two light chains forming a Y-shaped molecule that facilitates antigen binding.
Biological function summary

Immunoglobulin G1 plays a significant role in adaptive immunity as it can engage with different cellular receptors and complement proteins to mediate immune responses. It does not function as part of a complex by itself but rather interacts with other immune system components to execute its tasks. Mouse IgG1 is known for its ability to neutralize pathogens agglutinate invading microbes and opsonize them for enhanced phagocytosis by immune cells like macrophages and neutrophils.

Pathways

The signaling processes associated with mouse IgG1 involve pathways such as the Fc gamma receptor pathway and the classical complement pathway. Engagement of its Fc region with Fc gamma receptors on the surface of immune cells leads to effector functions like phagocytosis and antibody-dependent cellular cytotoxicity (ADCC). In these pathways proteins like complement component C1q and other Fc gamma receptor subtypes play key roles in modulating the immune responses initiated by IgG1.

Immunoglobulin G1 is relevant in conditions like autoimmune diseases and allergic reactions where inappropriate immune activation or regulation can be observed. In autoimmune diseases for example IgG1 can form immune complexes that deposit in tissues and activate inflammatory cascades involving proteins like complement factors and other immunoglobulin subclasses. Its involvement in allergic responses often involves IgG1 competing with IgE antibodies for antigen binding altering the allergic response dynamics.
