Customer kindly contacted us to determine the concetration of Anti-CD34 antibody [4H11[APG]] (FITC) so they may use the appropriate amount of isotype control in Flow Cytometry.

Thank you for contacting us.

Unfortunately we have not determined the exact concentration for this product. Before conjugation with FITC this product has a 1mg/ml concentration. When using isotype controls in Flow Cytometry keep in mind that it is recommended that concentrations are really antibody amount in excess. Therefore we would recommend using ab18435 Mouse IgG1 Isotype control at a concentration matching or exceeding 1mg/ml.

Abcam also carries other products essential for Flow Cytometry such as isotype controls. If testing live cells we also recommend cell viability controls such as 7-AAD or Propidium Iodide. Propidium Iodide (PI) binds to double-stranded DNA. PI cannot cross intact plasma membrane and therefore will only be present in DNA of cells where the plasma membrane has been compromised/ permeabilized. We do carry PI and offer a 7-AAD Cell-Mediated Cytotoxicity Assay Kit.

Yes, you are absolutely correct. I can confirm, that the non- conjugated equivalent of the ab86886 is the ab86885.

When using an unconjugated antibody I can however also recommend to use an unconjugated isotype control. We forexample the ab91353as an mouse IgG1 unconjugatedisotype control.

For the ab51387 I recommend to use an isotype control which is of the same isotype and conjugation. I can therefore not recommend to use the ab18435 for this antibody as a control.

We do currently not have the optimal control for ab51387, which is a mouse IgG1 conjugated to PE and TR in our catalogue. I have inquired with my colleagues whether we can add this for you. I will contact you again, as soon as I have received an answer.

However, the customer thinks it might not be suitable and ab18435 Mouse IgG1, kappa monoclonal [MOPC-21] (FITC) - Isotype control

The anti-CD4 antibody [RPA-T4] (FITC), ab86886, is a Mouse IgG1. The best isotype control for ab86886 would be a Mouse IgG1 FITC conjugated such as ab18435.

ab37356 would be a good isotype control for a Mouse polyclonal IgG FITC conjugated.
