Immunocytochemistry/ Immunofluorescence - FITC Anti-HA tag antibody [EPR22819-101] (AB316174)

ab316174 staining HA-tag in HEK-293T cells transfected with HA-tagged TATA-box-binding protein (bottom row) and non-transfected HEK293T cells (top row). The cells were fixed with 4% formaldehyde (10 min), permeabilised with 0.1% Triton x-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab316174 at 0.5 μ/ml (shown in green). ab195884, rabbit monoclonal to alpha Tubulin (Alexa Fluor® 647) antibody (shown in magenta) was used at 2 μg/ml for co-staining. Nuclear DNA was labelled with DAPI (shown in blue).

Image was acquired with a high-content analyser (Operetta CLS, Perkin Elmer) and a maximum intensity projection of confocal sections is shown.

This product also gave a positive signal in 100% methanol (5 min) fixed cells under the same testing conditions.

Binds to sialic acid-containing receptors on the cell surface, bringing about the attachment of the virus particle to the cell. This attachment induces virion internalization of about two third of the virus particles through clathrin-dependent endocytosis and about one third through a clathrin- and caveolin-independent pathway. Plays a major role in the determination of host range restriction and virulence. Class I viral fusion protein. Responsible for penetration of the virus into the cell cytoplasm by mediating the fusion of the membrane of the endocytosed virus particle with the endosomal membrane. Low pH in endosomes induces an irreversible conformational change in HA2, releasing the fusion hydrophobic peptide. Several trimers are required to form a competent fusion pore..
