    Feline calicivirus is a common viral disease that affects cats. It is characterized by upper respiratory symptoms, pneumonia, oral ulceration, and occasionally arthritis. It is a fairly mild flu-like condition and rarely causes serious complications. FCV is a tiny non-enveloped virus consisting of a 20 sided protein capsid (that fashions cup like dimples) surrounding a single strand of positive sense RNA.

Do you have any information on the epitope of FCV1-43?
We also want to know which strains could be available for the mAb.

Answer

I was able to obtain the following information:

Not only does this antibody bind the 63kDa capsid protein, but it has also recently been shown to specifically recognize the vp2 region by binding to protein expressed by a construct specifically encoding vp2. The epitope recognized by this antibody is believed to be both linear and containing no glycosylation sites.

We were not able to obtain an image of the blot though, or information about specificity for strains of FCV.

We purchased last year, I think almost one year ago.
When we bought it, we checked by WB immediately.
We got the same results which we had sent to you.
We were so disappointed with the antibody.
I dont think it has degraded.
Then we gave up the antibody to use.
This year, we really need WB data using anti-FCV antibody.
Therefore we recheck it again.
I want you to confirm our data by yourselves.
I appreciate if you let us know your results.
We have ordered ab93284, one week ago.
We are waiting for it.

I have forwarded your data to the lab and I have asked if there is any problem with the lot you have.

I am assuming the QC test that they did confirmed reactivity with FCV, and that the sample did not contain FBS or serum of any kind. In any case, I think the best approach now is to see what results you get with ab93284.

It is possible that the vial of ab33990 you have now was defective when you received it and that it will not detect FCV of any strain. On the other hand, it is specific for a single epitope on the capsid, so there is the possibility that the capsid you are working with does not contain this epitope, if there is capsid variation among FCV strains.
