WB with Hela cells sees multiple bands but none at 72 kDa, a weak at around 35 kDa (isoforms) used also siRNA (4 different sequences) to reduce signal, but saw no reduction at all, sees however biological phenotype with siRNA (but not in WB) Ab: 1/1000 block: 5% milk secondary: OK, works well
