    Factor XIII is activated by thrombin and calcium ion to a transglutaminase that catalyzes the formation of gamma-glutamyl-epsilon-lysine cross-links between fibrin chains, thus stabilizing the fibrin clot. Also cross-link alpha-2-plasmin inhibitor, or fibronectin, to the alpha chains of fibrin.

      ab4057 staining formalin fixed paraffin-embedded human bowel tumour tissue sections.  The sample was subjected to heat mediated antigen retrieval in citrate buffer (pH 6.0) and blocked with 10% srum for 1 hour at 24°C, prior to incubation with the primary antibody (diluted 1/100 in milk) for 12 hours at 4°C.  A biotinylated goat anti-rabbit antibody was used as the secondary.

    Thank you for your prompt reply. If you wouldn't mind, I have some more questions. I know that Factor VIII related antigen also stains endothelial cells. Can these 2 things be used interchangablely? It seems strange that we would be instructed to test Factor XIIIa on placenta. Is there something special about placental vessels?

    Thank you for your enquiry. Both proteins are involved in the coagulation cascade. Whether you can use the antibodies interchangeably depends on the experiments you are doing. Unfortunately we aren't experts in the field, so we couldn't tell you which one would be better to use in your particular situation. I am not sure why placenta is suggested as the positive control. It is possible that the protein shows higher expression in placental blood vessels than vessels in other tissues. If you would like me to enquire further about this matter, I would be happy to.     The positive control listed for this antibody is placenta. Which placental cells should I use? In addition, the localization is listed as cytoplasmic and nuclear. Which localization gives more sensitive staining?

    Thank you for your enquiry. Factor XIIIa is a coagulation protein present in the plasma. Staining is cytoplasmic. It should stain the endothelial cells of the blood vessels. It appears that the datasheet states that the cellular localization is cytoplasmic and secreted, not cytoplasmic and nuclear. I completely missed that, and I apologize. I hope this information helps.     No, i didn't try placenta due to the absence of this tissue in our laboratory. Placenta is also recommended as a positive control in the protocol provided by Novocastra. In addition to it, skin can be used as a positive control too since dermal dendritic cells are immunoreactive to Factor XIIIa AB (ref. Novocastra catalog) and upregulation of this cell type is well described in different inflammatory conditions. I hope this information is helpful

    Thank you for your response. This antibody has been tested for IHC using human placenta so it would be very important to run an experiment with tested positive control alongside with the samples. As you have mentioned in your e-mail, dendritic cells also express Factor XIIIa. However, the expressed levels of Factor XIIIa may be different from that in the placenta which usually shows very high levels of this protein. We have looked through our order record and actually the batch number you received was 77421. We have sold many vials form this batch without any problem. It may well be that the vial you received has become inactivated during shipping and/or storage. We can offer you a new vial free of charge.
