ERAS also known as Embryonic Stem Cell Ras (EC-Ras) is a member of the Ras protein family. It has a molecular weight of approximately 23 kDa. ERAS is mainly expressed in embryonic stem cells and specific cancer cells. It shares structural similarities with other Ras family proteins having a low GTPase activity that contributes to its role as a molecular switch in signaling pathways. Like other Ras proteins ERAS toggles between active GTP-bound and inactive GDP-bound forms which influences downstream signaling pathways.
Biological function summary

The ERAS protein influences cell proliferation survival and growth in tissues where it is expressed. It is not part of a larger protein complex but rather acts independently to stimulate the phosphoinositide 3-kinase (PI3K) pathway. This activation of the PI3K pathway leads to the downstream activation of Akt a critical effector in cellular growth and survival processes. By promoting these cellular functions ERAS contributes to maintaining the self-renewal properties of stem cells and influencing oncogenic processes in cancer cells.

Pathways

ERAS engages actively with the PI3K/Akt signaling pathway facilitating the survival and growth of cells. It directly interacts with the PI3K protein promoting cellular proliferation. Additionally ERAS has interactions with the MAPK/ERK pathway a well-known pathway linked to cell division and differentiation. The interaction between these pathways involves proteins like Raf and MEK which integrates growth signals coming from activated Ras proteins.

ERAS expression has been associated with certain types of cancers notably germ cell tumors and possibly breast cancer. Through its interaction with the PI3K/Akt signaling pathway ERAS contributes to the oncogenic properties observed in these cancers. Moreover its activities can influence the behavior of proteins such as PTEN and p53 which are well known for their tumor suppressor functions. The modulation of these pathways and interactions ultimately affect cellular processes like apoptosis which can contribute to the development and progression of cancer.
