    Binds activated protein C. Enhances protein C activation by the thrombin-thrombomodulin complex; plays a role in the protein C pathway controlling blood coagulation.

    Expressed strongly in the endothelial cells of arteries and veins in heart and lung, less intensely in capillaries in the lung and skin, and not at all in the endothelium of small vessels of the liver and kidney.

      Immunofluorescent analysis of 4% Paraformaldehyde fixed, 0.1% Triton X-100 permeabilized HUVEC (human umbilical vein endothelial cells) labelling EPCR/CD201 with ab300525 at 1/50 dilution followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed secondary antibody at 1/1000 (2 μg/ml) dilution (green).       Tubulin is detected with ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) (red) at 1/200 dilution.

      Flow Cytometry analysis of Jurkat (human T cell leukemia T lymphocyte, Left) / HUVEC (human umbilical vein endothelial cell, Right) labeling EPCR/CD201 (red) with ab300525 at a 1/500 dilution. A Goat Anti-Rabbit IgG (Alexa Fluor® 488, (ab150081)) was used as the secondary antibody at a 1/2000 dilution. Rabbit monoclonal IgG (ab172730) Isotype control (black). Unlabeled control - Cells without incubation with primary and secondary antibodies (blue).

      Gated on viable cells.

      Negative control: Jurkat (PMID: 11703343).

      EPCR/CD201 was immunoprecipitated from 0.35 mg HUVEC (human umbilical vein endothelial cell) whole cell lysate 10 ug with ab300525 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab300525 at 1/1000 dilution. VeriBlot for IP secondary antibody (HRP)(ab131366) was used at 1/1000 dilution.

      Lane 1: HUVEC (human umbilical vein endothelial cell) whole cell lysate 10 ug

      Lane 2: ab300525 IP in HUVEC whole cell lysate

      Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab300525 in HUVEC whole cell lysate

      Blocking and dilution buffer and concentration: 5% NFDM/TBST.

      Exposure time: 3 minutes
