This antibody is suppliedas ascites and not purified. Ascites contains other proteins in addition tothe specific antibody. Since the antibody concentration is usually measured by measuring protein content it is not possible to supply your customer with the antibody concentration.

We generally expect 5 to 10mg/ml of specific antibody in ascites.

Thank you very much for your inquiry. Indeed, this antibody is an ascite and the concentration has not been determined. The concentration is indeed difficult to determine, as an ascite is not a purified antibody and contains other proteins. the concentrations of an antibody in an ascite normally vary from 1 to 10mg/ml. In order to set up the ELISA, I do however not think that the concentration is necessary. Indeed, the antibodies should be used at saturating conditions to obtain reproducible results. This means, that when setting up the ELISA with different concentration of this antibody, a antibody dilution condition should be used where the values are well in the plateau of the curve (meaning well saturating the measure). In order to be able to create a standard curve and quantify therefore the protein of interest, a sample with the known concentration of the protein of interest (in this case Enterovirus 71 proteins) should be used. This can be either the recombinant proteins or a sample with know concentration. In the standard curve, the antibodies should be used in saturating conditions again and the only variable should be the concentration of the protein of interest. I hope this information was helpful. I have received the information from the laboratory that according to the originators, the clone completely neutralized the ability of the BrCr strain to infect cells (the originators indicate that they used RD and THP-1 cells) at a 1/14 dilution. Please find below the protocol to use ab36367: The fixation the we would recommend for the use of this product in IF is cold acetone for 10 minutes. The cells will not require further fixation and the recommended dilution is between 1/500 and 1/2000. To dilute the antibody use a proteinaceous buffer pH 7.5 to pH 8.0, eg PBS with 1% BSA.
