You are correct in thinking that the anti-Endothelin A Receptor antibody (ab85163) has been used successfully in staining frozen tissue sections, however, this has as yet only been performed in rat tissue. This does not mean that it will not work with the mouse tissue, simply that we have not tried it. The homology of the immunogen used (Synthetic peptide conjugated to KLH derived from within residues 100 - 200 of Rat Endothelin A Receptor, SwissProt reference P26684), shares very high homology (100%)with the mouse protein (SwissProt reference http://www.uniprot.org/uniprot/Q61614). We would therefore predict that the antibody would be able to detect the mouse protein. However as we have not tried it ourselves we are not able to guarantee this.

If you would be interested you would again be eligible for a testing discount if you wanted to try ab85163 in staining frozen sections of mouse tissue.

As we discussed, I would recommend permeabilizing the cells with 0.1-0.2% Tween 20 instead of with Triton, and using a 1:500-1:1000 dilution of antibody. Please keep me updated about the results using these alterations.

Regarding these antibodies, all of the information that is currently available can be found on the product datasheets on our website. I have summarized the answers to your questions below. a) All of these antibodies have been tested in WB and with human samples and are guaranteed to work. These antibodies have been tested in the following samples: ab76259 - LOVO, COS-7; ab84673 - HeLa, HepG2, MCF7; ab117521 - HEK293. Currently, no one has let us know that they have published with these antibodies, therefore no referencese are currently available. Our website will be updated should this information become available. Additionally, no specific protocol is recommended for these products. The recommended starting dilutions for WB are: ab76259 - 1ug/ml; ab84673 - 1ug/ml; ab117521 - 1:5000. b) Currently, we do not have a distributor, but sell directly to Greece. Orders do require prepayment by credit card. c) The pricing for each antibody is €370 plus €42 for shipping.
