Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-ELMO1 antibody (AB2239)
IHC image of ab2239 staining in human normal lymphoid formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab2239, 10µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
The protein ELMO1 participates in the regulation of Rac signaling pathways through the formation of complexes with DOCK family proteins. This complex formation contributes to the activation of Rac a small GTPase critical for actin cytoskeleton dynamics. ELMO1 affects processes such as phagocytosis and cell migration by modulating cytoskeletal configurations showing its role in the cellular machinery controlling cell shape and movement.
Pathways
The ELMO1 protein contributes to the Rac1 signaling and the phagocytosis pathway. These pathways play essential roles in integrating extracellular signals to promote cytoskeletal rearrangement and cellular movement.
