補足情報

ELL3 also known as Eleven-nineteen Lysine-rich Leukemia protein 3 has a molecular weight of around 70 kDa. It functions as an elongation factor belonging to the ELL family which includes ELL and ELL2. This protein exhibits expression in various tissues but higher levels occur in testes and several cancer cell types. ELL3 enhances the process of transcription elongation by positively regulating the rate at which RNA polymerase II progresses along the DNA template allowing efficient synthesis of full-length transcripts.
Biological function summary

The protein acts as a component of the super elongation complex (SEC) which suggests its role in maintaining active transcription in cells. ELL3 contributes to the regulation of gene expression important for cell development and proliferation. This protein influences the transcriptional output of genes associated with cell cycle control and differentiation. Through the SEC it interacts with several transcription factors and co-factors that integrate various signals for effective gene expression tuning.

Pathways

ELL3 plays an essential role in transcriptional pathways that involve RNA polymerase II. The protein is critical in the transcription elongation pathway which modulates the transition from transcription initiation to productive elongation. It interacts with proteins such as EAF1 and members of the AF4/FMR2 and MYC family demonstrating its interconnection within the transcriptional machinery and signaling networks that control cellular growth and energy metabolism.

ELL3 has associations with certain types of cancers particularly prostate cancer and breast cancer. Dysregulation or altered expression of ELL3 can contribute to oncogenic processes and tumor progression. In this context ELL3's interaction with proteins like ELL and ELL2 within cancer pathways emphasizes its role in cell proliferation and survival in malignant tissues. Understanding ELL3's involvement in these diseases aids in the potential development of targeted therapies for cancer treatment.

ターゲットの情報

Enhancer-binding elongation factor that specifically binds enhancers in embryonic stem cells (ES cells), marks them, and is required for their future activation during stem cell specification. Does not only bind to enhancer regions of active genes, but also marks the enhancers that are in a poised or inactive state in ES cells and is required for establishing proper RNA polymerase II occupancy at developmentally regulated genes in a cohesin-dependent manner. Probably required for priming developmentally regulated genes for later recruitment of the super elongation complex (SEC), for transcriptional activation during differentiation. Required for recruitment of P-TEFb within SEC during differentiation. Probably preloaded on germ cell chromatin, suggesting that it may prime gene activation by marking enhancers as early as in the germ cells. Promoting epithelial-mesenchymal transition (EMT) (By similarity). Elongation factor component of the super elongation complex (SEC), a complex required to increase the catalytic rate of RNA polymerase II transcription by suppressing transient pausing by the polymerase at multiple sites along the DNA. Component of the little elongation complex (LEC), a complex required to regulate small nuclear RNA (snRNA) gene transcription by RNA polymerase II and III (PubMed : 22195968).
