EGF stimulates the growth of various epidermal and epithelial tissues in vivo and in vitro and of some fibroblasts in cell culture. Magnesiotropic hormone that stimulates magnesium reabsorption in the renal distal convoluted tubule via engagement of EGFR and activation of the magnesium channel TRPM6. Can induce neurite outgrowth in motoneurons of the pond snail Lymnaea stagnalis in vitro (PubMed:10964941). -

Immunohistochemical analysis of paraffin-embedded Mouse kidney tissue labelling EGF with ab288294 at 1/1000 (0.5 µg/ml) followed by a ready to use LeicaDS9800 (BOND™ Polymer Refine Detection) was used. Positive staining on the distal convoluted tubules in mouse kidney. The section was incubated with ab288294 for 30 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin.
Secondary antibody only control: Secondary antibody is a ready to use LeicaDS9800 (BOND™ Polymer Refine Detection).

Exposure time: 3 minutesBlocking and diluting buffer and concentration are 5% NFDM/TBST.
Negative control: Mouse spleen and Rat spleen.
The molecular weight observed is consistent with what has been described in the literature (PMID:1636692, 2260661, 7499272 & 2268351).
Freshly made lysate is highly recommended to avoid EGF degradation and for better detection.

EGF was immunoprecipitated from 0.35 mg mouse kidney lysate with ab288294 at 1/30 dilution (2µg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using aab288294 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.
