Dux4 also known as Double homeobox 4 is a transcription factor with an important role in gene regulation. It has a molecular mass of approximately 52 kDa and is expressed mainly in germline and early embryonic tissues but it is usually silenced in adults. Dux4 contains a double homeobox domain which facilitates its ability to bind DNA sequences and regulate target gene expression. Despite its limited expression in adult tissues Dux4 is present in low levels in certain muscle tissues.
Biological function summary

Dux4 regulates a wide range of genes associated with muscle differentiation and apoptosis. It is not typically part of a known protein complex but acts independently to influence gene activity. Its function as a transcription factor impacts muscle cell differentiation by activating various myogenic and apoptotic genes. This regulatory activity plays a major role in normal muscle function and development in embryos.

Pathways

Dux4 plays a significant role in the Wnt signaling and beta-catenin pathways. These pathways are important for tissue development and cellular proliferation. Dux4 interacts with proteins like beta-catenin to influence cell growth and differentiation. Inappropriate activation of these pathways due to Dux4 involvement can result in dysregulated cell growth impacting muscle tissue integrity.

The inappropriate expression of Dux4 is related to facioscapulohumeral muscular dystrophy (FSHD) and cancer. FSHD involves the misexpression of Dux4 in muscle tissues leading to muscle weakness and degeneration. In cancer Dux4 misexpression can contribute to tumor development and progression by disrupting normal cell differentiation processes. Dux4's interaction with proteins involved in these conditions such as beta-catenin helps to elucidate its role in disease pathology.

Binds to double-stranded DNA elements with the consensus sequence 5'-TAATCTAATCA-3' (PubMed : 28459454, PubMed : 28459457, PubMed : 29572508, PubMed : 30315230, PubMed : 30540931). Binds to chromatin containing histone H3 acetylated at 'Lys-27' (H3K27ac) and promotes deacetylation of H3K27ac.
