DNA Polymerase iota also known as POLI is a specialized DNA polymerase involved in DNA repair processes. It is characterized by its ability to catalyze translesion synthesis permitting DNA replication across lesions that stall the replication fork. Mechanically POLI incorporates nucleotides opposite certain types of DNA damage albeit with higher error rates compared to other polymerases. It is encoded by the POLI gene which in humans is located on chromosome 18. The mass of the protein approximates 83kDa. DNA Polymerase iota is expressed primarily in the testis and is found in other tissues but in lower amounts. Its localization and activity underline its unique roles in maintaining genome stability under conditions that challenge the DNA replication machinery.
