DENTT also known as DEP domain-containing protein 5 (DEPDC5) is a member of the GATOR complex. It has a molecular mass of approximately 160 kDa. DENTT is expressed in various human tissues with high levels in the brain and skeletal muscle. This protein plays a role in the mechanisms of nutrient sensing and detecting energy levels within cells.
Biological function summary

DENTT regulates the activity of the mechanistic target of rapamycin complex 1 (mTORC1). As part of the GATOR1 complex it serves to inhibit mTORC1 in response to nutrient levels. By doing so DENTT participates in cellular processes such as growth and metabolism. Its activity is linked to the control of cell size and autophagy.

Pathways

DENTT is involved in the mTOR signaling pathway an important regulator of cell growth and proliferation. It interacts with proteins such as NPRL2 and NPRL3 which form part of the same GATOR1 complex. The interplay between DENTT and these proteins modulates mTORC1 activity aligning cellular metabolism with nutrient availability.

Alterations in DENTT are implicated in familial focal epilepsies. Mutations within the DEPDC5 gene can disturb mTOR signaling leading to imbalanced neuronal excitability. Additionally aberrant DENTT activity connects with proteins like RHEB known to drive mTORC1 activity which contributes to the pathogenesis of certain epileptic conditions.

Part of the CASK/TBR1/TSPYL2 transcriptional complex which modulates gene expression in response to neuronal synaptic activity, probably by facilitating nucleosome assembly. May inhibit cell proliferation by inducing p53-dependent CDKN1A expression.
