    I have some equiry about two antibodies, DE1 (ab41349) and DE2 (ab41696). Since, your technical specialist had replied me before that "we have tested them on recombinant DIII coated plates and they were not reactive." I would like to know what is the recombinant Domain III as your technical person mention? Can you give me more informations about the the recombinant Domain III? Also, do your technical person mean that both antibodies, DE1 (ab41349) and DE2 (ab41696), cannot attach on recombinant DIII coated plates?

    Thank you for your enquiry. Basically, the epitopes for DE1 and DE2 are complex, conformational epitopes and probably non-linear - and since the immunogen is not a peptide, the amino acid sequence is not known. The information on the recombinant DIII coated plate is just additional information. Both monoclonal antibodies work with native antigens and may work with recombinant proteins which are folded properly. It may be that our recombinant DIII has not folded perfectly and therefore, both monoclonal antibodies did not work with our recombinant DIII. Unfortunately, there is no further information on this failed testing. I am sorry I can't provide more information as there is no more info on the Domain III testing results.     Thank you for your enquiry. DE1 (ab41349) and DE2 (ab41696) are monoclonal antibodies from two different fusion experiments but the immunizing protocol is the same for both fusions. As mentioned in the datasheets, although they are both dengue group specific, DE1 is the monoclonal that is currently being used in the MAC-ELISA for detection of dengue-specific IgM. We believe that DE2 will function just as well in the MAC-ELISA but extensive comparisons have not been carried out. Additionally, we have tested them on recombinant DIII coated plates and they were not reactive. I hope this information will be useful.
