Cytokeratin 2 also known as KRT2 is a type II cytokeratin with a molecular weight of approximately 66 kDa. This protein is primarily expressed in the suprabasal layers of the epidermis especially in the upper spinous and granular layers of the skin. KRT2 plays a role in forming and maintaining the structural integrity of the epithelial cells due to its ability to assemble into intermediate filaments that provide mechanical support and resilience to epithelial tissues.
Biological function summary

Cytokeratin 2 contributes to epidermal differentiation and the formation of the skin barrier. It acts as part of the cytoskeletal network in keratinocytes providing structural support to the cells. Cytokeratin 2 is often found operating together with other keratin proteins as part of a complex. Its filamentous structure helps resist stress and external forces allowing the skin to withstand physical and chemical insults.

Pathways

Cytokeratin 2 has a significant role in keratinocyte differentiation and the skin barrier formation pathways. It is intricately related to other members of the keratin family such as Cytokeratin 1 (KRT1) which further regulate these processes. Cytokeratin 2 expression affects pathways that control cell proliferation and apoptosis within the skin ensuring the balance between cell division and cell death necessary for maintaining healthy skin tissue.

Deficiencies or mutations in Cytokeratin 2 can lead to Ichthyosis Bullosa of Siemens a condition characterized by blistering and peeling skin. Mutations may affect the stability and assembly of keratin filaments compromising skin integrity. Cytokeratin 2 abnormalities connect with other cytokeratin proteins like KRT1 which also contribute to similar skin disorders. Understanding its role and interactions with other proteins may assist in the development of treatments for keratin-associated skin diseases.
