CYP2S1 also known as Cytochrome P450 2S1 is a member of the cytochrome P450 superfamily of enzymes. This protein has a molecular weight of approximately 56 kDa. It is mainly expressed in extrahepatic tissues with high presence noted in the liver lungs and small intestine. Its expression is also observable in epithelial tissues and certain tumor types. The function of CYP2S1 involves oxidation reactions particularly those linked to fatty acids and xenobiotic metabolism.
Biological function summary

CYP2S1 plays an essential role in drug metabolism and the detoxification of chemicals. It participates in the processing of endogenous and exogenous compounds and operates alongside other members of the cytochrome P450 family. While CYP2S1 is not part of a conventional multi-protein complex its interactions with other proteins in the family influence its activity. Its reductase partners assist in electron transfer required for enzyme functionality.

Pathways

CYP2S1 is involved in pathways associated with xenobiotic metabolism and the degradation of specific fatty acids. It functions in these pathways by acting on substrates introduced by diet or environmental exposure. CYP1A1 and CYP2B6 are other cytochrome P450 enzymes that share interactions with CYP2S1 influencing shared metabolic and signaling pathways. This cooperation provides metabolism versatility and regulates substrate-specific detoxification.

CYP2S1 has connections to cancer and inflammatory conditions. It is especially relevant in the context of lung and breast cancer where altered expression levels are often observed. The enzymes CYP2E1 and CYP1A2 also participate in cancer-related processes due to their roles in the activation of pro-carcinogens. In inflammatory diseases the activity of CYP2S1 influences lipid mediator levels which can contribute to disease progression or suppression.

A cytochrome P450 monooxygenase involved in the metabolism of retinoids and eicosanoids (PubMed : 12711469, PubMed : 21068195). In epidermis, may contribute to the oxidative metabolism of all-trans-retinoic acid. For this activity, uses molecular oxygen inserting one oxygen atom into a substrate, and reducing the second into a water molecule, with two electrons provided by NADPH via cytochrome P450 reductase (NADPH--hemoprotein reductase) (PubMed : 12711469). Additionally, displays peroxidase and isomerase activities toward various oxygenated eicosanoids such as prostaglandin H2 (PGH2) and hydroperoxyeicosatetraenoates (HPETEs) (PubMed : 21068195). Independently of cytochrome P450 reductase, NADPH, and O2, catalyzes the breakdown of PGH2 to hydroxyheptadecatrienoic acid (HHT) and malondialdehyde (MDA), which is known to act as a mediator of DNA damage (PubMed : 21068195).
