Question

Thank you for the response. I have another question about your CKII antibodies. Among those listed on the abcam catalog, which one can detect both alpha and alpha prime subnits on Western blot and with high titer. I also like to know how much cell extract (amount of total protein) need to be loaded/lane for detection of CK11 on western blot. Best Regards

Thank you for your enquiry and patience. Under the conditions tested, which included WB detection at optimized dilutions using lysate from cells expressing endogenous amounts of the subunits, they were specific as specified. That is, ab10466, ab10468 and ab10470 react with the alpha subunit and ab10474 reacts with the alpha prime subunit. We would recommend loading 20-30 ug protein lysate. CKII exists as a tetramer and is composed of an alpha, an alpha-prime, and two beta subunits. It states that the immunogen used was a synthetic peptide, which represented a portion of human casein kinase 2, alpha 1 polypeptide encoded within exon 3. Does this antibody recognize the alpha subunit or the alpha-prime subunit?

Question

I am interested in using your antibodies generated against human CK2 alpha peptides on Drosophila samples. The immunogen is listed as exon 3 (ab10466), exon 12 & 13 (ab10468) or exon 14 (ab10470) of the human CK2 alpha protein. However, the genbank listing for human CK2 alpha only describes 8 total exons, so I am confused about what the exact immunogenic peptide sequences are. Could you please send me the amino acids corresponding to these immunogens, so that I may compare them to the Drosophila sequence and see which has the greatest chance of cross-reacting.

Thank you for your enquiry. The immunogen information for the products are as follows: Ab10466- The immunogen used lies between aa 1-50. Ab10468- The immunogen used lies between aa 300-350. Ab10470- The immunogen used lies between aa350-391. The Ab most likely to work in Drosophila is Ab10648 (although this has not been tested), the other two Abs will almost certainly not function against Drosophila.
