Anti-Cortisol 抗体 [XM210] (ab1949)

To our knowledge, ab1949 nor ab6066 have not yet been tested in detecting canine cortisol and testosterone. However, as cortisol and testosterone are steroids, antibodies raised against them should detect them regardless of the species that they are produced in as their structure and composition should be the same.

We have two antibodies directed against cortisol which have been used in ELISA previously. These are the mouse monolonals ab1949 and ab116600. However these have not been used in a sandwich ELISA format or "diurnal cortisol assay and subsequently a cross reactant assay" as you have described and we would therefore not be able to guarantee that they would be suitable for this experiment. If you could describe in a bit more detail how you intend to use the antibody I may be able to comment further on whether these antibodies are likely to work in this setting.

Alternatively we have ELISA kits designed to detect cortisol in human serum or plasma (ab108665), urine (ab108674) or saliva (ab154996) which may be of interest to you. These are based on a competitive immunoenzymatic colorimetric method intended for the quantitative determination of Cortisol.

The antibodies you have suggested for DHEA (ab54202, ab35062 and ab123987) should be fine for what the customer has requested.

The same is true of the anti-DHEA-S (ab31668). The anti-Cortisol antibodies ab1949 and ab116600 can also be suggested. The anti-Androstendione you have suggested should also be suitable (ab34944).

We do also have an antibody raised to target the hydroxy-Pregnenolone (ab34916). I am not sure if this is what the customer is after but it may be worth them having a look.

The secondary antibody they need would depend on which antibodies they choose to buy in the end. We ahve a very good search engine which you can use to find suitable antibodies:

All you need to do is put in the isotype of the primary antibody and the species they are raised in (available on the datasheet of the primary antibody), specify that it is to be used in ELISA and that it is an HRP conjugate you want.

To be honest we do not have an antibody that we have tested with this kind of novel assay, and therefore do not know if it would work nor can we guarantee it.

Although ab1949 may work, I may recommend ab1952: It is also a mouse monoclonal, is guaranteed to work in competitive binding assays (which might be more related to the intended experimental approach) and crucially detects also free cortisol. Furthermore, there is no cross-reactivity with cortisone, if this is of importance to the customer. (ab1951 is similar, but it cross-reacts with cortisone).

Also, we unfortunately do not have Cortisol-3-CMO-NHS ester or something similar available in our catalogue at the moment.

Thanks for your inquiry. Yes ab1949 should work in mouse serum. However, there will likely be an issue with non-specific binding of your secondary antibody, which will be an anti-mouse IgG, with the IgG in the mouse serum. Of course, this is assuming you are not doing a direct ELISA, which is probably the best way to test your sample to eliminate the secondary antibody issues. I hope this answers your question.
