Claudin-14 (CLDN14) plays an important role in the mechanics of epithelial tight junctions contributing to paracellular transport regulation. It forms a barrier that controls the flow of ions and small molecules between cells. CLDN14 has a molecular weight of around 23 kDa. It expresses mainly in the epithelial cells of various tissues including the kidney where it regulates calcium reabsorption and balance in the body.
Biological function summary

Claudin-14 functions as an integral component of tight junction complexes. It interacts with other claudins occludins and junctional adhesion molecules to maintain the paracellular permeability. This protein contributes to the formation and function of the tight junctional structures ultimately influencing ion selectivity and permeability. Studies have shown its role in maintaining electrolyte composition and osmotic balance in cellular environments.

Pathways

Claudin-14 integrates in the calcium-sensing pathway and the tight junction signaling pathway. It associates with calcium-sensing receptors (CaSR) in the kidneys impacting calcium homeostasis. Tight junction signaling involves interactions with proteins such as ZO-1 which is important for maintaining tight junction integrity and regulating epithelial barriers. These connections ensure the coordination of ion transport and stringent control of cellular environments.

Mutations or dysregulations in Claudin-14 link to familial hypomagnesemia with hypercalciuria and nephrocalcinosis (FHHNC) and sensorineural deafness. FHHNC results from impaired calcium reabsorption affecting kidney function and calcium balance. Sensorineural deafness arises due to disorder in ion homeostasis in the inner ear where Claudin-14 is also present. Its interactions with CaSR and other tight junction proteins highlight the importance of Claudin-14 in maintaining specific physiological balances related to these conditions.
