      Accessory protein for MHC class-II antigen/T-cell receptor interaction. May regulate T-cell activation. Induces the aggregation of lipid rafts.

      Cell membrane. Localizes to lipid rafts. Removed from plasma membrane by HIV-1 Nef protein that increases clathrin-dependent endocytosis of this antigen to target it to lysosomal degradation. Cell surface expression is also down-modulated by HIV-1 Envelope polyprotein gp160 that interacts with, and sequesters CD4 in the endoplasmic reticulum.

        Flow Cytometry - Anti-CD4 antibody [W3/25] (ab6413)

        Lewis rat splenocytes stained with ab6413 (right) or mouse IgG1κ (ab170190) isotype (left). Lewis rat splenocytes were incubated for 30 min on ice in 1x PBS / 10 % rat serum to block FC receptors and non-specific protein-protein interaction followed by the antibody (ab6413) or mouse IgG1κ (ab170190) (1x106 in 100 µl at 0.2 µg/ml) for 30 min on ice.

        The secondary antibody Goat anti-mouse IgG H&L (Alexa Fluor ® 488, pre-adsorbed) (ab150117) was used at 1/2000 dilution for 30 min on ice. The cells were simultaneously stained with CD3 antibody.

        Acquisition of >30,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter. Events were gated on viable lymphocytes.

      Regarding the use of ab6413 in IHC-formalin fixed paraffin embedded sections, a 1:400 dilution is recommended and I suggest incubating overnight at 4 C. Please have your customer refer to our IHC-Paraffin sections protocol which can be found on our website (click on the protocols tab located on the online datasheet for ab6413).       This antibody has not been tested in frozen tissue sections so we are not aware of its suitability. We do not routinely offer free or trial sized samples for testing purposes. Our policy at Abcam is that if an antibody does not work as specified on the datasheet, we will offer a replacement or reimbursement.       I purchased antibody to label active cells in lymphoid tissues (spleen, thymus, MLN). Used 5um sections of 10% NBF for 24 hr tissues routinely processed to paraffin. I followed your online IHC protocol for paraffin sections. I used the AR protocol for the pressure cooker. I ran the antibody at 1/50, 1/200 and 1/400. The secondary antibody was anti-Mouse antibody was from Vector diluted 1/150. The negative control was completely absent of staining as expected. However, the test slides were also negative except for three or four cells that had cytoplasmic staining, that looked liked dead cell debris. I am very optimisitic about purchasing since you are the only company that I could find that sells an anti-rat CD4 that claims to work on formalin fixed paraffin sections. Many thanks,

      This antibody has ben tested on paraffin sections so it should work nicely. Please have a look at the datasheet for references on staining and preparation of sections.
