    Receptor for the Fc region of complexed or aggregated immunoglobulins gamma. Low affinity receptor. Involved in a variety of effector and regulatory functions such as phagocytosis of immune complexes and modulation of antibody production by B-cells. Binding to this receptor results in down-modulation of previous state of cell activation triggered via antigen receptors on B-cells (BCR), T-cells (TCR) or via another Fc receptor. Isoform IIB1 fails to mediate endocytosis or phagocytosis. Isoform IIB2 does not trigger phagocytosis.

    Is the most broadly distributed Fc-gamma-receptor. Expressed in monocyte, neutrophils, macrophages, basophils, eosinophils, Langerhans cells, B-cells, platelets cells and placenta (endothelial cells). Not detected in natural killer cells.

    Note=A chromosomal aberration involving FCGR2B is found in a follicular lymphoma. Translocation t(1;22)(q22;q11). The translocation leads to the hyperexpression of the receptor. This may play a role in the tumor progression.

      This data was developed using AB290737, the same antibody clone in a different buffer formulation.

      Immunohistochemical analysis of 4% PFA-fixed, 0.2% Triton X-100 permeabilized frozen mouse spleen (fresh) tissue labeling CD32B with ab290737 at 1/100 (11.1 μg/ml) dilution followed by ab150113 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) at 1/1000 dilution (2 μg/ml) (Green). Positive staining on mouse spleen is observed. The nuclear counterstain was DAPI (Blue).

      Secondary antibody control:  PBS was used instead of primary antibody followed by secondary antibody ab150113 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488)at 1/1000 dilution (2 μg/ml).

      Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized mouse splenocytes cells labelling CD32B with AB290737 at 1/250 dilution (4.44 μg/ml), followed by ab150117 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 dilution (2 μg/mL) (Green). Confocal image showing membranous and cytoplasmic staining in subsets of mouse splenocytes is observed.

      ab179513 Anti-beta Tubulin rabbit monoclonal antibody was used to counterstain tubulin at 1/200  dilution (10 μg/ml) (Red). The Nuclear counterstain was DAPI (Blue).

      The negative controls are as follows: 

      -ve control 1: ab290737 at 1/250 dilution followed by ab150080 at 1/500 dilution.

      -ve control 2: ab179513 at 1/200 dilution followed by 150117 at 1/1000 dilution.

      Flow cytometric analysis of mouse peripheral blood mononuclear cells (PBMCs) cells labelling CD32B with AB290737 at 1/1000 dilution (0.1 μg)/ Right compared with a mouse monoclonal IgG / Left isotype control.  Goat anti-Mouse IgG (Alexa Fluor® 488, ab150113) at 1/5000 dilution was used as the secondary antibody. Cells were stained with mouse IgG or ab290737, then stained with rabbit anti-mouse CD19 (ab267394) conjugated to APC. The conjugation was performed by using APC Conjugation Kit - Lightning-Link® (ab201807). Gated on viable cells.

      Dot blot analysis of CD32B using AB290737 at 1:1000 (0.538 μg/ml) followed by a Peroxidase-Conjugated Goat anti-Mouse IgG (H+L) at 1:10,000 dilution.

      Exposure time: 3 minutes
