    Induces susceptibility to atherosclerosis (By similarity). Cell adhesion molecule which is required for leukocyte transendothelial migration (TEM) under most inflammatory conditions. Tyr-690 plays a critical role in TEM and is required for efficient trafficking of PECAM1 to and from the lateral border recycling compartment (LBRC) and is also essential for the LBRC membrane to be targeted around migrating leukocytes. Prevents phagocyte ingestion of closely apposed viable cells by transmitting 'detachment' signals, and changes function on apoptosis, promoting tethering of dying cells to phagocytes (the encounter of a viable cell with a phagocyte via the homophilic interaction of PECAM1 on both cell surfaces leads to the viable cell's active repulsion from the phagocyte. During apoptosis, the inside-out signaling of PECAM1 is somehow disabled so that the apoptotic cell does not actively reject the phagocyte anymore. The lack of this repulsion signal together with the interaction of the eat-me signals and their respective receptors causes the attachment of the apoptotic cell to the phagocyte, thus triggering the process of engulfment). Isoform Delta15 is unable to protect against apoptosis. Modulates BDKRB2 activation. Regulates bradykinin- and hyperosmotic shock-induced ERK1/2 activation in human umbilical cord vein cells (HUVEC).

    Expressed on platelets and leukocytes and is primarily concentrated at the borders between endothelial cells. Isoform Long predominates in all tissues examined. Isoform Delta12 is detected only in trachea. Isoform Delta14-15 is only detected in lung. Isoform Delta14 is detected in all tissues examined with the strongest expression in heart. Isoform Delta15 is expressed in brain, testis, ovary, cell surface of platelets, human umbilical vein endothelial cells (HUVECs), Jurkat T-cell leukemia, human erythroleukemia (HEL) and U937 histiocytic lymphoma cell lines (at protein level).

      Tumor vascularization and VEGF in ascites.

      (A) ID8, ID8-c-MYC, or ID8-KRAS cells (2 × 106) were intra-peritoneal injected into C57/BL6 mice. Mice were sacrificed when the body weight reached 23 g. Disseminations were obtained from mice when sacrificed. Tumor vascularization of each dissemination was assessed using CD31 immunohistochemistry (ab56299).

      Scale bars represent 200 μm and 50 μm at low and high magnification, respectively.

      Paraffin sections (4-μm-thick) of the biggest tumor sections from ID8, ID8-c-MYC, and ID8-KRAS mice were dewaxed in xylene and rehydrated through graded ethanol to water. Antigens were retrieved by boiling in 10 mM citrate buffer (pH 6.0) for 30 min. The cooled sections were incubated in a peroxidase blocking solution for 10 min to quench endogenous peroxidase. Sections were incubated in a protein blocking solution at room temperature for 10 min to block non-specific binding. Sections were then stained for Ki67 using rabbit monoclonal antibody against mouse Ki67, CD31 using a rat monoclonal antibody against mouse CD31 (ab56299, Abcam 1:100 dilution).

    Answer

    The antibody was raised against a mouse endothelial membrane preparation, and as it is a monoclonal antibody, it could target any region of the protein. The epitope has not been mapped, unfortunately, so we do not know whether it targets one of the termini or an internal region.

    On the bottom of the online datasheet for ab56299 there is a "Recommended IHC-P protocol with ab56299" link under the "Protocols" heading. In summation:

    1.Dewax and Rehydration: Place the slides in a rack and sequentially pass the rack through following solutions: Histoclear or Xylene twice, 10 minutes each; and then a graded series of 100%, 90%, 70%, and 50% ethanol, 2 minutes each.
    2. Wash the slides with tap water for 5 minutes and once with DI water.
    3. Antigen Retrieval: treat the slides with 20µg/ml proteinase K in PBS for 15-30 minutes at room temperature.
    4. Wash the slides with tap water for 5 minutes and quench the endogenous peroxidase treated in 3% H2O2 in DI water for 30 minutes
    5. Wash the slides with tap water for 5mins and soaking in PBS-Tween20 (0.1%)
    6. Block the slides with blocking buffer (2.5-25% BSA in PBS-Tween20) for 10 minutes
    7. Add primary antibody (Rat against mouse CD31 1:100) and incubate the antibody for 2 hours at room temperature, or for overnight at 4°C.
    8. Wash the slides with rotation in PBS-Tween20 for 3 times, 5 minutes each.
    9. Add anti rat secondary antibody and incubate at room temperature following manufacturer instructions.
    10. Wash the slides with rotation in PBS-Tween20 for 3 times, 5 minutes each.
    11. Add Polymer-HRP labeled anti rabbit (Dako: K4010) and incubate for 30 minutes at room temperature.
    12. Wash the slides with rotation in PBS-Tween20 for 3 times, 5 minutes each.
    13. Perform DAB Color development (reagents available in Dako K4010).. 15. Wash with tap water and count stain the nuclear with hematoxin.

    Question

    The customer did try their experiment with a single secondary antibody and only got weak signal. So they would like to try your suggested protocol and were wondering if your secondary rabbit anti-rat ab6703 would be suitable for that purpose.

    Question

    I have an enquiry about the “Recommended IHC-P protocol with ab56299 for Anti-CD31 antibody [RM0032-1D12]”, why do they recommend the use of 2 secondary antibodies (rabbit anti rat followed by HRP-anti-rabbit), instead of just HRP anti-rat.


    The second reference (in Abcam website): http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=20145133&dopt=Abstract - there, they use only one secondary antibody i.e. biotinylated goat anti-rat.


    The customer would like to know if they can use only one HRP-secondary antibody. They don’t have the 2 recommended secondary antibodies.

    Answer

    We recommend a double secondary antibody application to add an amplification step to increase the strength of the signal. You can use a single secondary step, but if they see a weak signal, then they may have to go to the double secondary method to get a stronger signal.
