Immunohistochemistry (PFA perfusion fixed frozen sections) - Anti-CACNA1A antibody (AB32642)

Immunofluorescent staining for CACNA1A using Rabbit polyclonal to CACNA1A (ab32642)in rat Purkinje (arrows) and granular (arrow heads) cells, of the rat cerebellum. Protocol details : Rats were intracardially perfused with 4% paraformaldehyde. Whole brain tissue was post-fixed overnight in the same fixative, and cryoprotected in 20% sucrose and frozen in OCT. 30µm coronal sections were cut by cyrostat for use in fre floating IHC. Primary antibody ab32642 was incubated overnight at 1/3000 at room temperature. Secondary antibody Alexa fluor 488 1/3000 was incubated for 2 hours at room temperature.

Western blot - Anti-CACNA1A antibody (AB32642)

This blot was produced using a 3-8% Tris Acetate gel under the TA buffer system. The gel was run at 150V for 60 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab32642 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.

補足情報

This supplementary information is collated from multiple sources and compiled automatically.

CACNA1A also known as Cav2.1 is a voltage-gated calcium channel subunit with an important role in calcium ion transport across cell membranes. It belongs to the P/Q-type calcium channels and weighs approximately 250 kDa. This protein is widely expressed in the central nervous system particularly in the cerebellum and cerebral cortex where it has an important function in neurotransmission and synaptic activity.
Biological function summary

CACNA1A influences synaptic transmission by modulating calcium ion entry into neurons which triggers neurotransmitter release. It forms a component of a larger heteromeric complex that involves other subunits which help regulate channel activity and stabilization. By facilitating calcium influx it supports activities such as muscle contraction and signal transduction in neuronal circuits.

Pathways

This calcium channel subunit participates in the calcium signaling pathway and synaptic vesicle cycle. Through these pathways CACNA1A interacts with proteins like CACNB and CACNA2D which modulate its channel functions. These interactions help in the rapid signal transmission required for effective synaptic communication and overall cellular homeostasis.

Mutations or dysregulation of CACNA1A can lead to conditions like familial hemiplegic migraine and episodic ataxia type 2. It has associations with proteins such as IRS4 which may influence the onset or progression of these neurological disorders. Understanding these interactions aids in elucidating the molecular basis of such conditions and potentially guiding therapeutic strategies.

ターゲットの情報

Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1A gives rise to P and/or Q-type calcium currents. P/Q-type calcium channels belong to the 'high-voltage activated' (HVA) group and are specifically blocked by the spider omega-agatoxin-IVA (AC P30288) (PubMed : 1311418). They are however insensitive to dihydropyridines (DHP).
