    Activation of C5 by a C5 convertase initiates the spontaneous assembly of the late complement components, C5-C9, into the membrane attack complex. C5b has a transient binding site for C6. The C5b-C6 complex is the foundation upon which the lytic complex is assembled.
    Derived from proteolytic degradation of complement C5, C5 anaphylatoxin is a mediator of local inflammatory process. It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes. C5a also stimulates the locomotion of polymorphonuclear leukocytes (chemokinesis) and direct their migration toward sites of inflammation (chemotaxis).

    An association study of C5 haplotypes and genotypes in individuals with chronic hepatitis C virus infection shows that individuals homozygous for the C5_1 haplotype have a significantly higher stage of liver fibrosis than individuals carrying at least 1 other allele (PubMed:15995705).

    I have been checking with my lab colleagues the percentage of homology of the epitopes recognised by our anti C5 antibodies and the correspondent protein in bovine species. Unfortunately, I am afraid none of our products is very likely to cross react with this species.

    I am looking for an antibody that binds both C5 and C5a and I came across ab11876 which looks very promising. In researching anti-C5/C5a antibodies, I noticed that clone #557 was cited in Klos et al. Journal of Immunological Methods, 111 (1988): 241-252. The ab11876 antibody data sheet identifies this antibody as derived from clone 557. Can you confirm that ab11876 is the same clone 557 that is identified in the Klos et al. paper? Also, I need an anti-C5/C5a antibody that binds both C5 and C5a, does not inhibit the activation of C5 and does not prevent the activation or inhibit the activity of C5b. Can you confirm that the ab11876 antibody exhibits all these characteristics? Many Thanks,

Nevertheless we can confirm the clone mentioned in this article is the same as our product. The ab11876 recognizes an epitope located within the alpha-chain of C5 that corresponds to C5a. Thus this product recognizes both C5 and C5a. It inhibits binding of C5a to the C5a receptor through a competitive mechanism similar as described in Kola et al, Immunotechnology, 1996, 2:115-126.
