    C4 plays a central role in the activation of the classical pathway of the complement system. It is processed by activated C1 which removes from the alpha chain the C4a anaphylatoxin. The remaining alpha chain fragment C4b is the major activation product and is an essential subunit of the C3 convertase (C4b2a) and the C5 convertase (C3bC4b2a) enzymes of the classical complement pathway.
    Derived from proteolytic degradation of complement C4, C4a anaphylatoxin is a mediator of local inflammatory process. It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes.

    Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-C4d antibody (ab87424)
    IHC image of C4d staining in human liver carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab87424, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    Additional bands at: 110 kDa, 130 kDa, 30 kDa, 48 kDa. We are unsure as to the identity of these extra bands.


    Exposure time: 30 seconds


    C4d circulates in blood as a disulfide-linked trimer of an alpha, beta and gamma chain. Our immunogen sequence is within the alpha chain. The band at 85kDa could correspond to the alpha chain.

    ICC image of ab87424 stained HepG2 cells. The cells were 100% Methanol fixed (5 min) and then incubated in 1%BSA / 10% normal Goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab87424, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 Goat anti-Rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM. This antibody also gave a positive result in 100% Methanol fixed (5 min) HeLa, and MCF-7 cells at 5µg/ml.

Question

Dear Miss/Mr,
C4d is a split product derived from C4 and is generated after complemet activation. Complement activation only occurs extracelulary. Therefore C4d ONLY can be generated extracellulary and covalently deposited on cell surfaces. It is truth that hepatocytes produce C4, the native protein, but C4d only can be deposited on the SURFACE of cells. There is plenty of basic literature about it. My question is whether your antibody recognizes the native form of C4 or only the split product C4d (neoantigen). In the case of the antibody specifically recognizes C4d and not C4, is this molecule internalized? Thank you very much for all your help. Yours sincerely,

Answer

Thank you for your inquiry.

The immungens for these antibodies correspond to sequences within the C4d. This will also lead to the recognition of the complement precursor that is intra cellular since the complement precursor also contains theC4d sequence.

In a healthy sample there should be not complement deposited on cell surfaces. The lab did not test disease models and therefore we do not have an image of C4d that is deposited on cell surfaces.

We will update the on-line datasheet for this product as soon as we receive additional data.

Inquiry: Dear Miss/Mr, I am thinking about buying the anti-C4d antibodies (ab87424) and (ab64157) for immunohistochemistry. In these websites: https://www.abcam.com/C4d-antibody-ab64157.html#C4d-Primary-antibodies-ab64157-3.jpeg https://www.abcam.com/C4d-antibody-ab87424.html#C4d-Primary-antibodies-ab87424-3.jpg you show pictures of a hepatocarcinoma stained for C4d with both antibodies. However, as shown in the pictures, the staining is found mainly in the cytoplasm. C4d is complement fragment deposited on cell membrane after complement activation. My question is why C4d staining shown in the pictures is cytoplasmatic? Is an artefact? Is C4d internalized? On that case do you have any reference to confirm that C4d is internalized? Thank you for your inquiry.

The liver (mainly hepatocytes) is responsible for biosynthesis of 90% of plasma complement components and their soluble regulators, including the classical (C1r/s, C2, C4, C4bp), alternative (C3, factor B), lectin (MBL, MASP1-3, Map19), terminal (C5, C6, C8, C9) pathways of the complement system, and soluble regulators (factors I, H, and C1 inhibitor).

The images shown on the datasheets of ab87424 and ab64157 are of liver samples. Since these cell produce the complement they will show it in cytoplasma. The secreted complement will be washed away in IHC and ICC and therefore cannot be detected.

Complement is in the blood stream andis supposed to be deposited on bacteria or other invading microorganisms. In some disease like lupus etcthe tightly regulatedsystem gets disregulated and therefore complement deposits can be found on body cells. I suggest to review the available literature to get more information.
