    • 機能

      C3 plays a central role in the activation of the complement system. Its processing by C3 convertase is the central reaction in both classical and alternative complement pathways. After activation C3b can bind covalently, via its reactive thioester, to cell surface carbohydrates or immune aggregates.
      Derived from proteolytic degradation of complement C3, C3a anaphylatoxin is a mediator of local inflammatory process. In chronic inflammation, acts as a chemoattractant for neutrophils (By similarity). It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes.
      C3-beta-c: Acts as a chemoattractant for neutrophils in chronic inflammation.
      Acylation stimulating protein: adipogenic hormone that stimulates triglyceride (TG) synthesis and glucose transport in adipocytes, regulating fat storage and playing a role in postprandial TG clearance. Appears to stimulate TG synthesis via activation of the PLC, MAPK and AKT signaling pathways. Ligand for C5AR2. Promotes the phosphorylation, ARRB2-mediated internalization and recycling of C5AR2 (PubMed:8376604, PubMed:2909530, PubMed:9059512, PubMed:10432298, PubMed:15833747, PubMed:16333141, PubMed:19615750).

      Increased levels of C3 and its cleavage product ASP, are associated with obesity, diabetes and coronary heart disease. Short-term endurance training reduces baseline ASP levels and subsequently fat storage.

      C3b is rapidly split in two positions by factor I and a cofactor to form iC3b (inactivated C3b) and C3f which is released. Other proteases produce other fragments such as C3d or C3g. C3a is further processed by carboxypeptidases to release the C-terminal arginine residue generating the acylation stimulating protein (ASP). Levels of ASP are increased in adipocytes in the postprandial period and by insulin and dietary chylomicrons.
      Phosphorylated by FAM20C in the extracellular medium.

      We have a customer who is experiencing the following problem: ?I am writing in relation to a C3 primary antibody (mouse monoclonal ab11874) I just purchased through abcam just a few weeks ago. I am running some westerns with serum and liver samples and am finding it very difficult to obtain clear bands using the C3 antibody. There are often no bands present on the gel at all!!! Do you have any recommendations for the primary and secondary antibody concentrations?? Your quick reply would be much appreciated?.

      Thank you for your enquiry and I'm sorry to hear that your customer is experiencing difficulty with this antibody. Without knowing more details about what your customer has already tried, I would suggest trying the primary at 1:500 and optimizing from there based on results. To increase the signal, make sure to load at least 20-30 ug of lysate and incubate with the primary overnight at 4C. Is the customer using an Abcam secondary antibody? If the customer is still having trouble with the antibody, please send us their detailed protocol (go to the online datasheet and use the complaint link in the box under application notes).

      I am working with cardiovascular research lab of university of Berne (Switzerland). We are going to create our own complement ELISA (for C4d and C3a) to analyse human plasma samples. My question is: could you give me an advise which of your products can be used as antibody-pairs for the mentioned purpose?

      The antibodies our source has in their product range gave only one option for setting up an immunoassay to quantify complement C3a, but this option will also recognize C3. The option is: ab11874: monoclonal antibody to human C3/C3a (C-terminus, clone 2898) in combination with ab11872: monoclonal antibody to human C/C3a (clone 474). We know that ab11874 and ab11872 are useful as coating antibody. The usefulness of these antibodies as tracer antibody is only observed with C3 protein used as coating. However we expect that both antibodies are also useful as tracer antibody. If you are interested in these antibodies we are able to provide you with special antibodies without albumin (to use as coating and for labeling). In return we hope you are able to provide us with the test results.
