Western blot - Anti-BNIP3L/NIX (phospho S81) antibody [EPR19826] - BSA and Azide free (AB251494)

This data was developed using ab208190, the same antibody clone in a different buffer formulation.

The molecular weight observed is consistent with what has been described in the literature (PMID : 10467396).

The plasmids were kindly provided by our collaborator Dr. Zhong Chen, Zhejiang University.

Blocking and dilution buffer : 5% NFDM/TBST.

Exposure time : 3 seconds.

All lanes:

Western blot - Anti-BNIP3L/NIX (phospho S81) antibody [EPR19826] (<a href='/products/primary-antibodies/bnip3l-nix-phospho-s81-antibody-epr19826-ab208190'>ab208190</a>) at 1/5000 dilution

Lane 1:

HEK-293 (human embryonic kidney) transfected with an empty vector (vector control) containing a DDDDK tag, whole cell lysate at 10 µg

Lane 2:

HEK-293 transfected with a DDDDK-tagged mouse BNIP3L/NIX (WT) expression vector, whole cell lysate at 10 µg

Lane 3:

HEK-293 transfected with a DDDDK-tagged mouse BNIP3L/NIX (WT) expression vector, lysate then treated with alkaline phosphatase for 1 hour, whole cell lysate at 10 µg

Lane 4:

HEK-293 transfected with a DDDDK-tagged mouse BNIP3L/NIX S81A mutant expression vector, whole cell lysate at 10 µg

Induces apoptosis. Interacts with viral and cellular anti-apoptosis proteins. Can overcome the suppressors BCL-2 and BCL-XL, although high levels of BCL-XL expression will inhibit apoptosis. Inhibits apoptosis induced by BNIP3. Involved in mitochondrial quality control via its interaction with SPATA18/MIEAP : in response to mitochondrial damage, participates in mitochondrial protein catabolic process (also named MALM) leading to the degradation of damaged proteins inside mitochondria. The physical interaction of SPATA18/MIEAP, BNIP3 and BNIP3L/NIX at the mitochondrial outer membrane regulates the opening of a pore in the mitochondrial double membrane in order to mediate the translocation of lysosomal proteins from the cytoplasm to the mitochondrial matrix (By similarity). May function as a tumor suppressor (By similarity).
