    Toxoplasma is a crescent shaped sporozoan that lives as an intracellular parasite in various tissues of many vertebrates and completes its life cycle in a single host. Its life cycle includes two phases called the intestinal (or enteroepithelial) and extraintestinal phases. The intestinal phase produces oocysts and occurs only in cats, wild as well as domesticated. The extraintestinal phase occurs in all infected animals including cats, and produces tachyzoites (actively proliferating trophozoites) and eventually, bradyzoites (slowly growing trophozoites) or zoitocysts. Infection due to Toxoplasma gondii occurs in pregnant women where a variable degree of immunosuppression may exist or in patients receiving immunosuppressive drug therapy. Toxoplasma infects tissue of the GI tract where an active infection is accompanied by fever and enlargement of the spleen. Symptoms of toxoplasmosis are generally mild but severe infection of lymph nodes may occur. Congenital toxoplasmosis, in which the maternal infection is transmitted during pregnancy, can produce blindness or mental retardation in the newborn.

    If you are looking for an antibody to detect Toxoplasma gondii in frozen tissue sections, then the best choice is probably ab15170 or ab23507. Even though neither have been tested on sections of frozen tissue, the IHC data for detection in paraffin-embedded tissue is very clean and specific, for ab15170, and detection in frozen tissue is generally easier than detection in formalin-fixed paraffin-embedded tissue. The alternative, goat polyclonal ab23507, has been published twice.

    For detection of ab15170, you will need an anti-rabbit IgG secondary antibody conjugated to an enzyme such as HRP, or a fluorochrome such as FITC. For an HRP conjugate, we recommend ab6802:

    1- I will select a Primary antibody [(Anti-Toxoplasma gondii antibody (Biotin) (ab69251)], I wonder if the secondary antibody that I most to choose [(Rabbit polyclonal Secondary Antibody to Goat IgG - H&L (HRP) (ab6741)] is work with the primary antibody [(Anti-Toxoplasma gondii antibody (Biotin) (ab69251)].

    2- Would you please advise me which kind of buffer and the dilution factor I can use to dilute the primary as well as the secondary antibodies for ELISA test.

    The secondary antibody ab6741 will probably recognize the biotinylated antibody ab69251 but they have not been tested together. You may want to consider using the streptavidin-HRP conjugate ab7403 instead of a secondary antibody. Or, you may want to order a different primary antibody, without biotin, such as the HRP-conjugated ab21182:

    The Toxoplasma gondii antibody ab69251 should be used at a dilution of 1/1000 but you may need to optimize the dilution factor depending on your results. I recommend diluting the antibody in Tris-buffered saline, pH 7.4 for detection. The secondary ab6741 can be applied very dilute at 1/75,000 but again, this may need to optimized since these two antibodies have not been used together before.
