S.aureus binds IgG molecules by their Fc region. In serum, the bacteria will bind IgG molecules in the wrong orientation on their surface, which hinders opsonization and phagocytosis. Mutants of S. aureus lacking protein A are more efficiently phagocytosed in vitro, and mutants in infection models have diminished virulence. Due to its affinity for the Fc region of many mammalian immunoglobulins, protein A is considered a universal reagent in biochemistry and immunology.

    Question

    Dear technical team,

Our customer has some problem with abcam's antibodies.
Her application is similar ELISA but, not guaranteed one so, she needs to check ELISA test to prove antibodies' activity.
    She has below antibodies.
    https://www.abcam.com/Staphylococcus-aureus-antibody-Biotin-ab68954.html
    https://www.abcam.com/Staphylococcus-aureus-antibody-Biotin-ab35192.html
    https://www.abcam.com/Staphylococcus-aureus-antibody-702-ab20002.html
She purchased live staphylococcus to test ELISA but,
I'm not sure ifit can be used inindirect ELISA.
Can itbe coated on the plate?
Please give her a good idea about test method.

    Best regards,

    Answer

    Thank you for contacting us.

The anti-Staphylococcus aureus antibodies ab68954, ab35192 and ab20002 have not all been tested in ELISA and cannot therefore be guaranteed in this application. Ab35192 has currently only been used in ICC/IF but this does not mean that it would not work in this application, simply that we have not tested it and do not know if it will be suitable.

With the Anti-Staphylococcus aureus antibody [702] (ab20002), the antibody was used in a direct ELISA protocol using the heat killed Salmonella organisms directly to the plate. The cells were not lysed prior to coating on the plate. The Staph. aureus cells (5 x 107/ml) were bound to the wells of Immulon II 96-well plates in 50ul of 100 mM carbonate buffer, pH 9.6, per well. The plates are sealed with plastic wrap and incubated overnight at room temperature. The next day, the coating solution is removed by inverting the plates. The wellswere then blocked for 30 minutes with 1% BSA-PBS-0.05% Tween 20. This solution was then removed by inverting the plates. The coated plates are then ready for testing with the antibody.

I hope this information has been of help. If I can be of any further assistance, please do not hesitate to contact us again.

    Answer

    Thank you for contacting us.

Please find below the information I managed to collect by contacting the sources of the Anti-Staphylococcus aureusand the Anti-Streptococcus agalactiaeantibodies we have in our catalog :

    Anti-Staphylococcus aureus:
https://www.abcam.com/ab23568 shows no reactivity to any streptococcus sp., mycoplasma sp. or corynebacteria sp.

Unfortunately, ab35192 has not been specifically tested against streptococcus so we are unable to guarantee it won’t react to S. agalactiae.

ab68950, ab68952, ab68954: These Staphylococcus aureus polyclonal antibodies are not absorbed for related microorganisms and so therefore could cross react with related bacteria such as Staph. Epidermidis.

MLS : Staphylococcus aureus: We do not have an antibody that is specific to Staph. aureus but does not have crossreactivity to Staph. epidermidis.

    Anti-Streptococcus agalactiae:
https://www.abcam.com/ab21126 has weak crossreactivity with Strep. dysgalactiae and does not cross-react with Strep. uberis, Strep. bovis, Strep. faecalis, Staph. aureus, Mycoplasma bovis, Mycoplasma californicum, Mycoplasma bovigenitalium, E. coli or Corynebacterium bovis.

I hope this information is helpful to you. I am still waiting for more information about ab128711 (Anti-Streptococcus agalactiae antibody [224/46]).
Please do not hesitate to contact us if you need any more advice or information.

    Question

Dear Sir/Madam I found 2 possibilities but they are not indicated for immunoprecipitation. Do you think that the antibodies ab68954 or ab20920 can be used for this assay? Thank you in advance for your information.

    Answer

Thank you for contacting us. Indeed, neither ab68954 nor ab20920 have been tested for immunoprecipitation (IP) yet, but they could theoretically be used in this application. If it is just between the two, I would recommend ab20920 because it is an unconjugated antibody. The biotin-labeled antibody ab68954 might not bind to the Protein A or G agarose as well as the unconjugated antibody. Also, ab20920 seems to detect its antigen(s) in a rather native state (as it works with frozen tissue sections) which might be more suitable for IP. However, these antibodies are polyclonal antibodies against various soluble and structural antigens of the whole Staphylococcus aureus. By using the antibodies in IP, a variety of proteins would be pulled down. If this is not appropriate for your experimental setting, I would be pleased to assist you in finding a suitable antibody (for example against a certain target protein). If you decide to go ahead and purchase one of these products though, you might be eligible for our testing discount program. With this, you get a discount off a future purchase if you buy the antibody now, test it in IP and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of 1 free primary antibody. The Terms and Conditions of this offer can be found at: www.abcam.com/collaborationdiscount I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

    Question

Hi, I would like to know whether this biotin-conjugated antibody ab68954 attaches onto a certain domain on the cell surface of (intact) S. aureus 27660, or whether the bacterial cell membrane has to be perforated and the antibody has to be internalized for it to have a binding reaction? If the binding is on the cell surface, what domain or receptor does the antibody attach to? Our application requires that we have the bacterial cells intact, and we need an antibody against staph that will conjugate on the cell surface. Thank you,

    Asked on Dec 01 2011

    Answer

Thank you for contacting us with your question, and for your patience while I have been in touch with the lab about your enquiry. My contact at the lab has informed me that the antibody was raised against a mixture of intact and lysed bacteria. No particular antigens were identified as targets. The antibody can recognize intact, bacterial cell surface proteins as well as internal proteins within permeabilized bacteria. I hope this information will be useful, but if you have any further questions or if there is anything else that we can do for you please do let me know.

    Question

P {margin-top:0;margin-bottom:0;} Dear Abcam, I've purchased your Rabbit polyclonal Anti-Staphylococcus aureus antibody (Biotin), ab68954 and I use it for IHC.n Recently, i got worrie about the unspecific background staining, therefore I look for a suitable isotype control. Would you perhaps recommend any control for this antibody? Sincerely yours

    Answer

Thank you for your inquiry. As a isotype control I would recommend general rabbit IgG that is biotin conjugated and tested for IHC. Unfortunately, we do not offer this specific isotype control at the moment, but only un-conjugated rabbit IgG (ab27478, Rabbit polyclonal IgG). I would like to recommend checking the Biocompare website which has an excellent antibody search facility that includes many suppliers. The links are: www.biocompare.com http://www.biocompare.com/ProductCategories/2045/Antibodies.html I am sorry to hear you have had difficulty obtaining satisfactory results from this antibody. We will happily offer technical support and in the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund. This is our Abpromise. If you would like me to investigate this particular case further for you, please reply to this email. I have attached a questionnaire, which will help me get a good overview of the protocol that has been used. I hope this information is helpful to you. Please do not hesitate to contact me if you have any further questions in this regard.
