Immunocytochemistry - Anti-Bid Cleavage Site antibody (AB10640)
A549 cells stained for BID cleave site (green) using ab10640 at 1/250 dilution in ICC/IF. It was followed by Alexa Fluor 488 Goat Anti-Rabbit IgG Secondary Antibody at 1/400 dilution for 30 minutes at room temperature (Panel a). Nuclei (Panel b : blue) were stained with SlowFade® Gold Antifade Mountant DAPI. F-actin (Panel c : red) was stained with Alexa Fluor 594 Phalloidin. Panel d is a merged image showing cytoplasmic localization. Panel e shows no primary antibody control.

Biological function summary
Bid functions as a critical mediator of apoptosis. It acts within the intrinsic pathway a vital part of programmed cell death. Bid does not function as a monomer but can form complexes with other Bcl-2 family proteins such as Bax and Bak promoting mitochondrial outer membrane permeabilization. Upon cleavage tBid translocates to the mitochondria to facilitate the release of cytochrome c initiating the cascade of apoptotic events.
Pathways
Bid plays an integral role in the apoptotic signaling pathways. The intrinsic or mitochondrial pathway is the primary context in which Bid operates. This pathway involves several key proteins including Bax and Bak which cooperate with tBid to permeabilize the mitochondrial membrane. In addition to its role in apoptosis Bid is indirectly linked with the extrinsic apoptotic pathway through its interaction with caspase-8 which also induces its cleavage into tBid.
