A549 cells stained for BID cleave site (green) using ab10640 at 1/250 dilution in ICC/IF. It was followed by Alexa Fluor 488 Goat Anti-Rabbit IgG Secondary Antibody at 1/400 dilution for 30 minutes at room temperature (Panel a). Nuclei (Panel b : blue) were stained with SlowFade® Gold Antifade Mountant DAPI. F-actin (Panel c : red) was stained with Alexa Fluor 594 Phalloidin. BH3 interacting domain death agonist (BID) is a pro-apoptotic member of the Bcl 2 family. BID interacts with both Bcl 2 and Bax through its BH3 domain. It usually exists in an inactive form in the cytosolic fraction of living cells and becomes cleaved and activated by caspase 8 in response to TNF alpha or Fas ligand. Once BID is cleaved, the C-terminal 15 kDa fragment of BID (p15) translocates onto mitochondria and is sufficient to trigger cytochrome c release, resulting in cell apoptosis. BID serves as a direct molecular link between caspase 8 activation and mitochondrial death machinery.

The Bid Cleavage Site is a region on the Bid protein which is also known as BH3 interacting domain death agonist. This protein is part of the Bcl-2 family and acts as a pro-apoptotic member. The molecular weight of the full-length Bid protein is approximately 22 kDa. Bid is expressed in various tissues within the body including the heart and liver. During apoptosis caspases cleave Bid resulting in a truncated form called tBid which significantly enhances its pro-apoptotic activity.
Biological function summary

Bid functions as a critical mediator of apoptosis. It acts within the intrinsic pathway a vital part of programmed cell death. Bid does not function as a monomer but can form complexes with other Bcl-2 family proteins such as Bax and Bak promoting mitochondrial outer membrane permeabilization. Upon cleavage tBid translocates to the mitochondria to facilitate the release of cytochrome c initiating the cascade of apoptotic events.

Pathways

Bid plays an integral role in the apoptotic signaling pathways. The intrinsic or mitochondrial pathway is the primary context in which Bid operates. This pathway involves several key proteins including Bax and Bak which cooperate with tBid to permeabilize the mitochondrial membrane. In addition to its role in apoptosis Bid is indirectly linked with the extrinsic apoptotic pathway through its interaction with caspase-8 which also induces its cleavage into tBid.

Bid has shown significant implications in cancer and neurodegenerative diseases. In cancer altered Bid expression can contribute to resistance against apoptotic stimuli allowing cancer cells to survive and proliferate unchecked. In neurodegenerative diseases such as Alzheimer's dysregulation of Bid and its interactions with other apoptotic proteins can lead to excessive neuronal cell death. These processes involve interactions with proteins like Bcl-2 which often act as antagonists to Bid's pro-apoptotic functions impacting disease progression.
