Avidin is a glycoprotein that strongly binds to biotin and is expressed in the oviducts of birds reptiles and amphibians as well as throughout the egg whites of bird eggs. It has a molecular mass of about 66-69 kilodaltons and can exist in the form of monomer dimer or tetramer. Avidin can be used in different forms like avidin-HRP (Horseradish Peroxidase) conjugate and avidin peroxidase in various biochemical applications. Due to its high affinity for biotin avidin is quite popular in various purification and labeling tasks.
Biological function summary

Avidin binds biotin with high specificity and affinity forming one of the strongest known non-covalent interactions. Avidin is not usually part of a cellular complex but functions independently as a biotin-binding molecule. It plays a role in the regulation of biotin availability which is a critical cofactor for carboxylase reactions. The availability of biotin affects various metabolic processes since biotin is necessary for enzymes that play roles in gluconeogenesis fatty acid synthesis and the catabolism of branched-chain amino acids.

Pathways

The strong avidin-biotin binding is exploited in pathways related to signal amplification and detection. Avidin often in combination with peroxidase conjugates enhances the sensitivity and precision in biotechnological applications like ELISA and Western blot through its interaction with biotinylated molecules. Avidin's interaction with biotinylated chemical species allows cleaning and enrichment of biotinylated proteins from biological samples. Though avidin itself is not directly involved in cellular pathways the avidin-biotin system is instrumental for tagging and detecting molecular interactions.

Avidin does not have a direct disease association but serves a supportive role in research and diagnostic settings. The avidin-biotin system aids in the detection of biomarkers associated with cancer and infectious diseases such as monitoring biotin-labeled antibodies for the presence of specific antigens. There is no known association of avidin with specific proteins related to diseases but in laboratory settings it is used to enhance the detection and isolation of target molecules that are related to various conditions being studied.
