Question

Thank you for your quick reply. Indeed there are two isoforms of the
Aspartate aminotransferase (also referred to as Glutamate oxaloacetate
transaminase), one being localized in the cytosol, namly GOT1
(http://www.uniprot.org/uniprot/P17174) and one being localized in the
mitochondria, namely GOT2 (http://www.uniprot.org/uniprot/P00505).
However, the problem of a specific detection is the high similarity of
both isoforms. I have attached a alignment of both proteins in order
to visualise the high similarity. I came to the conclusion that this
was also the problem with antibodies from other companies, as they
detected both isoforms and were not able to distingush between the
specific isoforms, however they claimed their antibodies to be specific.

Thank you for your email. My colleague has passed this inquiry to me as she is busy today.

I have checked the sequence similarity between the two forms (http://www.ebi.ac.uk/Tools/services/web_clustalw2/toolresult.ebi?tool=clustalw2&jobId=clustalw2-I20130110-122843-0890-68773436-pg) which is quite low. So I am assuming that antibody ab98228, ab65941 and ab85857 could be a choice for you.

Please note, these antibodies are not tested for specificity toward mitochondiral AAT1 so the assumption is totally based on the low homology between sequences.

Dear Sir or Madam, I am searching for two specific antibodies against the cytosolic form (GOT1) and the mitochondrial form of the human aspartate aminotransferase. I have already tested antibodies, which claimed to be specific, from another company, however so far I could not find antibodies which could specifically detect one of the isoforms. Maybe you have already received this request from other costumers. Thank you for your help in advance.

We have indeed three antibodies which target GOT1. However, after checking the SwissProt database, I cannot find any information about different isoforms. Could you please go into more detail why the other antibodies from different companies were not suitable for you, so I can check if we have the right product for you in store?
