Immunocytochemistry/ Immunofluorescence - APC Anti-Cytokeratin 19 antibody [EP1580Y] (AB224980)
Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized MCF7 (human breast adenocarcinoma epithelial cell) cells labelling Cytokeratin 19 with ab224980 at 1 : 50 dilution (Magenta).
Confocal image showing cytoplasmic staining in MCF7 cell line (shown in magenta). The counterstain was observed in green. Nuclear DNA was labelled with DAPI (shown in blue). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
Negative control : SH-SY5Y.
ab195887 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 488) was used to counterstain tubulin at 1/200 dilution (Green).

Flow Cytometry (Intracellular) - APC Anti-Cytokeratin 19 antibody [EP1580Y] (AB224980)
Overlay histogram showing HeLa cells stained with ab224980 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Triton X-100 for 15 min. The cells were then incubated in 1x PBS / 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (ab224980, 1/5000 dilution) for 30 min at 22°C.
Isotype control antibody (black line) was Rabbit IgG (monoclonal) Allophycocyanin used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.
Acquisition of >5,000 events were collected using a 40 mW Red laser (640nm) and 670/14 bandpass filter.

Biological function summary
CK19 plays a role in maintaining the cytoskeletal framework that provides mechanical support and helps control cell size and shape. It is not part of a large protein complex but works closely with other cytokeratins to ensure the stability and resilience of epithelial tissues. The A53-B/A22.26 positive staining is often used to confirm its presence in clinical histopathology.
Pathways
CK19 connects to cellular processes especially involving the keratinization and epithelial cell differentiation pathways. It interacts with other cytokeratins including BA16 and KR17 to facilitate these processes. This interaction aids in modulating the dynamics of cytoskeletal rearrangement during cell movement division and differentiation.
