ANKLE1 also known as Ankyrin Repeat and LEM Domain containing 1 or LEM3 is a protein that functions as a DNA endonuclease. This protein has a molecular mass of approximately 63 kDa. It is expressed in various tissues with presence reported in the testis ovaries placenta and to a lesser extent in other tissues. ANKLE1 contains ankyrin repeat domains which often mediate protein-protein interactions. Through its mechanical action it is involved in DNA damage response.

Processes chromatin bridges during cytokinesis by cleaving DNA, both priming TREX1-mediated resolution and directly resolving bridges (PubMed : 36825683). Proper processing of chromatin bridges prevents their breakage during cytokinesis, thereby reducing DNA damage, micronuclei formation, and the activation of immune responses such as the cGAS-STING pathway (PubMed : 36825683).
