Alexa Fluor® 647 Anti-NKG2A 抗体 [EPR23737-127] (ab300745)

Immunohistochemical analysis of paraffin-embedded human tonsil labeling NKG2A with ab300745 at 1/100 dilution (5 µg/ml) (shown in red).The tissue section was first incubated with TBS containing 0.025% (v/v) Triton X-100, 0.3 M glycine and 1% (w/v) BSA for 1 h at room temperature to block non-specific protein-protein interactions, followed by overnight incubation at +4°C with ab300745 at 1/100 dilution in TBS containing 0.025% (v/v), Triton X-100 and 1% (w/v) BSA.  Nuclear DNA was labeled with DAPI (shown in blue). The section was then mounted using Fluoromount^®.  Positive membrane staining was observed on human tonsil tissue.

Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Heat mediated antigen retrieval was performed using ab93684 (Tris/EDTA buffer, pH 9.0).

Flow Cytometry (Intracellular) analysis of human peripheral blood mononuclear cells (PBMC) labeling NKG2A (right) with ab300745 at a 1/500 dilution.  Isotype control - Rabbit monoclonal isotype control - Alexa Fluor^® 647 (left).  Cells were stained with anti-CD56 conjugated to PE, fixed with 2% PFA for 10 min followed by intracellularly staining with rabbit IgG or ab300745.
