Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 488 Anti-PHOX2B antibody [EPR14423] - C-terminal (AB210459)
ab210459 staining PHOX2B in SKNSH cells. The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab210459 at 1/50 dilution (shown in green) and ab195889, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 594), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Biological function summary
PHOX2B influences the development of noradrenergic neurons and parasympathetic ganglia. It often functions as part of a transcription factor complex interacting with other proteins to drive the expression of genes required for the autonomic nervous system. The control PHOX2B exerts over neuronal differentiation makes it essential for maintaining the proper functioning of neurons responsible for involuntary bodily processes.
Pathways
PHOX2B is integral to the development and maintenance of the autonomic nervous and respiratory systems. It interacts closely with RET a receptor tyrosine kinase within the neurotrophic signaling pathways that regulate neuronal growth and survival. Additionally the protein plays a role in the endothelin signaling pathway further demonstrating its importance in neurodevelopment.
