Aflatoxin B1 (AFB1) is a potent mycotoxin produced by Aspergillus species particularly Aspergillus flavus and Aspergillus parasiticus. It is a small toxic compound with a molecular mass of about 312 grams per mole. Aflatoxin B1 commonly appears in agricultural environments as these fungi contaminate crops like nuts cereals and maize. In biological matrices the molecule often requires detection via sophisticated methods such as aflatoxin ELISA kits which facilitate sensitive and specific measurement of AFB1 levels.
Biological function summary

Aflatoxin B1 exerts its toxic effects by interacting with DNA and inducing mutations that can lead to cancer. It interacts with the cytochrome P450 enzyme complex in the liver where it undergoes bioactivation to form an epoxide. This reactive metabolite can covalently bind to DNA resulting in genotoxic stress. It does not form part of any large protein complexes on its own but relies on enzymatic conversion to exert its biological activity.

Pathways

AFB1 primarily involves the cytochrome P450 enzymatic system. This pathway activates AFB1 into a reactive epoxide linking it to the metabolic pathways of xenobiotic degradation. It is closely related to other cytochrome P450 enzymes such as CYP1A2 and CYP3A4 which also participate in metabolic activation and detoxification of various xenobiotics and drugs influencing their biological impact.

Aflatoxin B1 is strongly associated with hepatocellular carcinoma a primary form of liver cancer due to its genotoxic properties. It is also linked to acute aflatoxicosis symptoms of which include liver damage and impaired immunity. The protein p53 is commonly associated with AFB1-related liver cancer as mutations in p53 are facilitated by aflatoxin-induced DNA damage undermining the cell's repair mechanisms and contributing to oncogenesis.
