Western blot - Anti-ACE2 antibody (AB15348)
False colour image of Western blot : Anti-ACE2 antibody staining at 1/1000 dilution, shown in green; Mouse anti-GAPDH antibody [6C5] (ab8245) loading control staining at 1/20000 dilution, shown in red. In Western blot, ab15348 was shown to bind specifically to ACE2. A band was observed at 120-135 kDa in wild-type Caco-2 cell lysates with no signal observed at this size in ACE2 knockout cell line ab277338 (knockout cell lysate ab275516). To generate this image, wild-type and ACE2 knockout Caco-2 cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 5 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

Target Information
ACE2 (angiotensin-converting enzyme 2) is a crucial receptor for the SARS-CoV-2 virus, facilitating its entry into host cells and contributing to COVID-19 pathogenesis. Beyond COVID-19, ACE2 is involved in regulating the renin-angiotensin system, impacting blood pressure and inflammation, and is being explored as a therapeutic target for various infectious diseases. For instance, ACE2 has been implicated in the pathogenesis of SARS (Severe Acute Respiratory Syndrome) caused by the SARS-CoV virus, which also uses ACE2 as a receptor to enter host cells. Additionally, ACE2 expression and activity are being studied in the context of other viral infections, such as influenza and certain types of viral pneumonia, where it may influence disease severity and outcomes

Biological function summary
The ACE2 protein plays an important role in the regulation of cardiovascular and renal functions. It is a single-pass type I membrane protein and its activity reduces inflammation and oxidative stress in cells. ACE2 does not function as part of a larger protein complex but its enzymatic conversion has a substantial impact on reducing the effects of angiotensin II in the body leading to vasodilation and decreased blood pressure.
Pathways
ACE2 involvement is significant in the renin-angiotensin system and the kallikrein-kinin system. These pathways are essential for maintaining cardiovascular homeostasis. In the renin-angiotensin system ACE2 works in opposition to angiotensin-converting enzyme (ACE) balancing the effects through the production of angiotensin-(1-7) from angiotensin II. Additionally ACE2 interacts indirectly with proteins like angiotensin receptor type 1 (AT1) and angiotensin receptor type 2 (AT2) ensuring proper signaling and physiological responses.
