Interpolated concentrations of spike IL-1 beta in rat serum plasma and cell culture media with 10% fetal bovine serum samples.

The concentrations of IL-1 beta were measured in duplicates interpolated from the IL-1 beta standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50% plasma (citrate) 50% plasma (EDTA) 50% and 1640 RPMI media 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2).

How the assay works

Rat IL-1 beta ELISA Kit, Chemiluminescent (ab327306) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min

Potent pro-inflammatory cytokine. Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. Promotes Th17 differentiation of T-cells. Synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells. Plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6. Involved in transduction of inflammation downstream of pyroptosis : its mature form is specifically released in the extracellular milieu by passing through the gasdermin-D (GSDMD) pore.
