Abcam's beta NGF Rat ELISA Kit (ab193736) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of rat beta NGF in cell lysate and tissue lysate.

This assay employs an antibody specific for rat beta NGF coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures beta NGF present in the samples. The wells are washed and biotinylated anti-rat beta NGF antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of beta NGF bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Beta nerve growth factor (beta-NGF) is a significant protein that functions in the development and maintenance of the nervous system. It is also known as NGF or NGFB and has a molecular mass of around 26 kDa. This protein is highly expressed in a wide range of tissues including the brain skin and various glands. Beta nerve growth factor plays an important role in the growth survival and differentiation of neurons. As a member of the neurotrophin family it binds to receptors like TrkA and p75NTR on the surface of nerve cells. Nerve growth factor is important for the development and maintenance of the sympathetic and sensory nervous systems. Extracellular ligand for the NTRK1 and NGFR receptors, activates cellular signaling cascades to regulate neuronal proliferation, differentiation and survival (By similarity). The immature NGF precursor (proNGF) functions as a ligand for the heterodimeric receptor formed by SORCS2 and NGFR, and activates cellular signaling cascades that lead to inactivation of RAC1 and/or RAC2, reorganization of the actin cytoskeleton and neuronal growth cone collapse. In contrast to mature NGF, the precursor form (proNGF) promotes neuronal apoptosis (in vitro) (By similarity). Inhibits metalloproteinase-dependent proteolysis of platelet glycoprotein VI (By similarity). Binds lysophosphatidylinositol and lysophosphatidylserine between the two chains of the homodimer. The lipid-bound form promotes histamine relase from mast cells, contrary to the lipid-free form (By similarity).
