Induction of p38 MAPKα (pT180/Y182) phosphorylation in HeLa cells in response to anisomycin treatment.
HeLa cells were cultured in 96-well tissue culture plates and treated (15 min) with a dose-range of anisomycin before cell lysis. Data from quadruplicate measurements of p38 MAPKα (pT180/Y182) are plotted and compared against Total p38 MAPKα protein levels.

p38 MAPK alpha ELISA Kit ab221012 is a rapid single-wash 90-min Other Immunoassay to measure p38 MAPK alpha in cell lysate, tissue homogenate. This SimpleStep sensitivity is 300 pg/mL.
How the assay works
p38 MAPK alpha SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. p38 MAPK alpha ELISA Kit ab221012 protocol summary
1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Biological function summary
P38 is involved in several cellular processes such as inflammation cell differentiation and apoptosis. It often functions as part of a MAPK signaling complex where it serves a critical role in transmitting signals from the cell surface to the nucleus. It interacts with upstream kinases for activation and affects cellular responses by phosphorylating transcription factors and other protein kinases. Through experiments using techniques like p38 western blot and alpha ELISA scientists can monitor its activity and understand its role in cellular physiology.
Pathways
P38 signaling is integral to both the MAPK and NF-kB pathways. It helps mediate several cellular responses including inflammation and stress responses. Within these pathways p38 interacts with other proteins such as JNK and ERK which helps regulate adaptive and innate immune responses.
