Interpolated concentration of native Uromodulin was measured in duplicate at different sample concentrations. Undiluted samples are 25% serum, plasma (citrate), plasma (EDTA) and plasma (heparin). The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Interpolated concentrations of native Uromodulin in mouse kidney tissue extract based on a 100 µg/mL extract load. The concentrations of Uromodulin were measured in duplicate and interpolated from the Uromodulin standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Uromodulin concentration was determined to be 41624 pg/mL in kidney tissue extract.

Mouse Uromodulin ELISA Kit, Chemiluminescent (ab325735 ) is a rapid single-wash sandwich 55-min ELISA to measure Mouse Uromodulin in Serum, EDTA Plasma, Urine, Cell culture media, Tissue Extracts, Citrate plasma with a sensitivity of 0.02 ng/mL.

How the assay works

Mouse Uromodulin ELISA Kit, Chemiluminescent (ab325735 ) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Mouse Uromodulin (ab325735 ) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature

Uromodulin. Functions in biogenesis and organization of the apical membrane of epithelial cells of the thick ascending limb of Henle's loop (TALH), where it promotes formation of complex filamentous gel-like structure that may play a role in the water barrier permeability. May serve as a receptor for binding and endocytosis of cytokines (IL-1, IL-2) and TNF. Facilitates neutrophil migration across renal epithelia.. Uromodulin, secreted form. In the urine, may contribute to colloid osmotic pressure, retards passage of positively charged electrolytes and inhibits formation of liquid containing supersaturated salts and subsequent formation of salt crystals (PubMed : 14871399, PubMed : 15327412). Protects against urinary tract infections by binding to type 1 fimbriated E.coli (PubMed : 11134021). Binds to the bacterial adhesin fimH which mediates the stable formation of bacterial aggregates, prevents the binding of E.coli to uroplakins UPK1A and UPK1B which act as urothelial receptors for type I fimbriae, and allows for pathogen clearance through micturation (By similarity). Also promotes aggregation of other bacteria including K.pneumoniae, P.aeruginosa and S.mitis and so may also protect against other uropathogens (By similarity).
