      Abcam’s Tweak Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse Tweak in serum, plasma and cell culture supernatants.

      This assay employs an antibody specific for Tweak coated on a 96-well plate. Standards and samples are pipetted into the wells and Tweak present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse Tweak antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Tweak bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    Nous avons effectué des test sur des surnageants, extraits cellulaires et bien entendu sur votre gamme étalon.

    A notre grande surprise nous avons obtenu un bruit de fond important pour le blanc ( > 0.500 de DO à 450 nm ) et aucune différence significative pour les différentes dilutions de la gamme étalon. Nous sommes particulièrement ennuyé pour nos expériences et pour l'investissement réalisé. Je vous joins un fichier excell qui vous montre les données obtenues.

    Suite à mon dernier email, j’ai eu un supplément d’informations du laboratoire. Voir les pièces jointes.

    Notez aussi que le standard ne doit pas être vortexer pour être mélanger mais plutôt solution C devrait être brièvement centrifugé avant l'ouverture puis une fois le diluant ajouté, la solution doit être mélangé en pipetant doucement la solution.

    Avez-vous brièvement vortexer « item c » avant utilisation ? Est-ce que la poudre a été bien dissoute ?
    Combien de fois avez-vous lavé la plaque ?
    Avez-vous ouvert la plaque juste avant utilisation ?
    Est-ce que les ODs ont été déterminés juste après que le « stop solution » a été ajouté ?

    I am sorry this item G of ab113320 did not containthe volumeas stated on the datasheet and for the inconvenience this has caused. As requested, I have issued a free of charge replacement with the order number xxx for 1 vial item G. As soon as we receive the vial it will ship to you - most likely early next week. The estimated delivery date is set for xxx.

    The vialwill be the new 200ul of 200X concentration, thus pleasedilute the component accordingly. Please alsospin the vialprior to opening to ensure complete recovery of the liquid at the bottomof the vial.

    I will be arranging for sending you 1 vial of item G (200ul, 200x). The process will be taking about a week and I expect that you will receive it next week.

    The online datasheet states incorrectly 240x, but the online protocol is correct in stating 200x. I'll have the datasheet information updated as quickly as possible. The volume of 200 ul is correct, though. I apologize about this confusion.

    This ELISA kit has not been tested against monomeric TWEAK/TNFSF12. It is only known to detect mouse TWEAK in its trimeric form. It is important to note that TWEAK is a homotrimer, meaning that all of the three units of the peptide are identical. Therefore, it is likely that the antibodies will recognize the monomers of TWEAK, but it is still unknown.
