Comparison of Bax distribution in subcellular fractions derived from 3.7x103 HeLa cells and whole cells cultured in the presence (treated) or absence (untreated) of 1 µM staurosporine for 4 hours.

Cells were collected directly after treatment and subcellular fractions were prepared using a cell fractionation kit (ab109719). The concentrations of Bax were measured in three different dilutions of the fraction samples in duplicate and interpolated from the Bax standard curve. The interpolated values are plotted (mean +/- SD, n=3). The mean Bax concentration was determined to be 841.4 pg/mL in the untreated cytosol fraction, 100.0 pg/mL in the untreated mitochondrial fraction, 912.6 pg/mL in the untreated whole cell sample, 186.9 pg/mL in the treated cytosol fraction, 1,366 pg/mL in the treated mitochondrial fraction, and 1,622 pg/mL in the treated whole cell sample.

Interpolated concentrations of native Bax in mouse cell extracts.

Interpolated concentrations of native Bax in mouse C2C12 cell extract based on a 12.5 μg/mL extract load, A20 cell extract based on a 5 μg/mL extract load, and PC-12 cell extract based on a 25 μg/mL extract load. The concentrations of Bax were measured in duplicate and interpolated from the Bax standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Bax concentration was determined to be 3,001 pg/mL in C2C12 cell extract, 1,404 pg/mL in A20 cell extract, and 1,689 pg/mL in PC-12 cell extract.

Other species reactivity was determined by measuring Bax in rat and human cell extracts within the working range of the assay, interpolating the protein concentrations from the mouse standard curve.

The mean Bax concentration was determined to be 243.5 ng/mg in C2C12 cell extract, 65.15 ng/mg in PC-12 cell extract, and 39.85 ng/mg in HeLa cell extract.

Bax also known as Bcl-2-associated X protein plays an important role in the regulation of apoptosis. It is widely known for its pro-apoptotic function opposing the action of anti-apoptotic proteins like Bcl-2. Bax has a molecular weight of approximately 21 kDa making it identifiable in laboratory techniques such as western blot. Researchers often use anti-Bax antibodies to detect and analyze its expression levels in various studies. The Bax protein is mainly expressed in mitochondria where it undergoes changes in response to apoptotic signals.

Biological function summary

The Bax protein forms homodimers or heterodimers with other Bcl-2 family proteins to facilitate apoptosis. This protein is part of a larger complex involving several members of the Bcl-2 family which finely tunes the balance between cell survival and programmed cell death. Through a change in its conformation Bax translocates to the outer mitochondrial membrane where it promotes the release of cytochrome c and other apoptogenic factors into the cytosol.

Pathways

Bax plays a significant role in the intrinsic pathway of apoptosis also known as the mitochondrial pathway. This pathway involves the release of cytochrome c which further activates downstream proteins such as caspase-9 and caspase-3. Bax interacts closely with proteins like Bak another pro-apoptotic member of the Bcl-2 family providing a cooperative function in mitochondrial membrane permeabilization. Together Bax and Bak act as gatekeepers of the mitochondrial pathway determining the cell's fate in response to apoptotic stimuli.

The dysregulation of Bax often associates with various cancers and neurodegenerative diseases. In cancer reduced Bax expression or function can lead to resistance to apoptosis contributing to unchecked cellular proliferation. Conversely in neurodegenerative disorders such as Alzheimer's disease increased Bax activity results in excessive neuronal apoptosis. The balance between Bax and its partner proteins like Bcl-2 or Bcl-xL is important in maintaining normal cellular function and preventing disease progression.

Accelerates programmed cell death by binding to, and antagonizing the apoptosis repressor BCL2 or its adenovirus homolog E1B 19k protein. Under stress conditions, undergoes a conformation change that causes translocation to the mitochondrion membrane, leading to the release of cytochrome c that then triggers apoptosis. Promotes activation of CASP3, and thereby apoptosis. BAX deficiency leads to lymphoid hyperplasia and male sterility, because of the cessation of sperm production.
