    Abcam’s TIMP4 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human TIMP4 in serum, plasma (collect plasma using EDTA and citrate as an anticoagulant. Heparin is not recommended), cell culture supernatants.


    This assay employs an antibody specific for Human TIMP4 coated on a 96- well plate. Standards and samples are pipetted into the wells and TIMP4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human TIMP4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TIMP4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Question

Dear Sirs,



We have collections of plasma samples from patients which we would like to assay

for certain cytokines and complement fractions.

However we noticed that certain manufacturers advice not to use these samples since heparin could interfere with the results of the assay.

Could you please explain which ELISA kits are suitable for analysis and which are not?



For the following Abcam product heparin plasma is not recommended:

Could you explain what are the reasons for not recommending the use with heparin samples? What concentrations have been assayed?



We need to evaluate if we need to re-take patient samples with other anti-coagulants or if heparinase treatment could allow us to assay our samples.

Thank you for your inquiry. Unofrtunately there is no straightforward way to categorize which kits can be used with heparin and which cannot. You need to look at the datasheet or contact us regarding the specific kit, as different manufacturers and reagents may or may not be suitable with heparin.

The reason for not using heparin is fairly straightforward though. When we tested heparin plasma, the detection of TIMP-4 was significantly decreased by some sort of interference cause by the heparin. EDTA and citrate plasma had no such interference.
