Sandwich ELISA - Human NT-proBNP ELISA Kit, Chemiluminescent (AB326386)

Serum of nine individual healthy human male donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean NT-proBNP concentration of detected samples was determined to be 10.9 pg/mL with a range of undetected – 20.6 pg/mL.

Interpolated concentrations of native NT-proBNP in human congestive heart failure serum samples.

The concentrations of NT-proBNP were measured in duplicates interpolated from the NT-proBNP standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 7.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean NT-proBNP concentration was determined to be 16839 pg/mL in serum.

Human NT-proBNP ELISA Kit, Chemiluminescent (ab326386) is a rapid single-wash sandwich 55-min ELISA to measure Human NT-proBNP in Serum, EDTA Plasma, Heparin Plasma, Citrate plasma, Cell culture media with a sensitivity of 0.82 pg/mL.

How the assay works

Human NT-proBNP ELISA Kit, Chemiluminescent (ab326386) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human NT-proBNP (ab326386) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection

Brain natriuretic peptide 32. Cardiac hormone that plays a key role in mediating cardio-renal homeostasis (PubMed : 1672777, PubMed : 17372040, PubMed : 1914098, PubMed : 9458824). May also function as a paracrine antifibrotic factor in the heart (By similarity). Acts by specifically binding and stimulating NPR1 to produce cGMP, which in turn activates effector proteins that drive various biological responses (PubMed : 1672777, PubMed : 17349887, PubMed : 17372040, PubMed : 21098034, PubMed : 25339504, PubMed : 9458824). Involved in regulating the extracellular fluid volume and maintaining the fluid-electrolyte balance through natriuresis, diuresis, vasorelaxation, and inhibition of renin and aldosterone secretion (PubMed : 1914098, PubMed : 9458824). Binds the clearance receptor NPR3 (PubMed : 16870210).. NT-proBNP. May affect cardio-renal homeostasis (PubMed : 17372040). Able to promote the production of cGMP although its potency is very low compared to brain natriuretic peptide 32 (PubMed : 17372040).. BNP(3-32).
